The growth and metastasis of tumour correlate to angiogenesis intimately,this complex process contains adherion,degradation of base material and migration.We can inhibit the tumorous invasion metastasis by inhibiting any part of this process in abstracto. Therefore, exploitation angiogenesis inhibitor is an athletic domain. Now anti-angiogenesis therapy has become the focus of cancer research.In 1970′Science Dr.Judah Folkman hypothesized that angiogenesis was obligatory for tumor growth. A tumor is unable to grow above 2mm3 and then towards into the dormancy period without development of a new blood vessel. When angiogenesis stimulators is stronger than angiogenesis inhibitors, the switch of angiogenesis is on and increases angiogenesis, which result in the proliferation,invasion and metastasis of tumor. Angiogenesis is known to be a complex process that requires endothelial cell proliferation,migration and differentiation into tube-like structures and controlled by extensive interactions between a various peptides and other modulating factors produced by host or tumor cell. But the research on traditional Chinese medicine targeted to angiogenesis just starts in our country.Systematic solvent separation method and column separation method was employed to extract different parts of Scutellaria barbatae D.DON. The effects of different parts of Scutellaria barbatae D.DON on proliferation of tumor cell were tested by MTT method. Select the most effective part and investigate the mechanism of inhibiting cancer growth and metastasis through apoptosis,tube formation experiment,immunofluorescence,ELISA,Griess assay.The main result and conclusion showed as below:1.Abstraction of Scutellaria barbatae D.DON:to abstract Scutellaria barbatae D.DON by column chromatography which useed mixed solvent CHCl3- MeOH and thin-layer chromatography (TLC),we found 7 different polarity parts,they were A-G;2.Experiment of different parts of Scutellaria barbatae D.DON selection: we found the crude extract of Scutellaria barbatae D.DON flavonoid group can inhibit proliferation of ECV-304 and tumour cells in contrast to the control group by MTT;3.The tumor cell apoptosis detected by FCM::Treated by different concentration of flavonoid(0μg/ml,30μg/ml,60μg/ml,120μg/ml and 240μg/ml),the rate of apoptosis increaseed,in contrast to the control group(P<0.01) with dose-effect correlation;4.The flavonoid group can depress the adherion capability of endothelial cell and tumor cell to basement membrane:we find the 40μg/ml concentration can showed signigicant effect of depress the adherion capability in contrast to the control group(P<0.01);5.The flavonoid group can depress the adherion capability of endothelial cell between tumor cell: After treated with different concentration of flavonoid 48 hours, the adherion capability decreased significantly, in contrast to the control group(P<0.01);6.Elisa assay:VEGF expression in SGC-7901 cell was markly inhibited by 20μg/ml,40μg/ml and 80μg/ml concentration of flavonoid group after 24 and 48 hours in contrast to the control group(P<0.01), but there were no significant differences between groups in 24 hours. MMP-2 expression in SGC-7901 cell was markly inhibited by 40μg/ml and 80μg/ml concentration of flavonoid group after 48 hours in contrast to the control group (P<0.01). With the same concentration, there are the significant differences in the inhibition of VEGF expression by 40μg/ml and 80μg/ml concentration between 24h and 48h (P<0.01). There are no significant differences between 20ug/ml flavonoid group and the control group.Conclusion: The flavonoid of Scutellaria barbatae D.DON can inhibit tumor metastasis and depress the adherion capability of endothelial cell and tumor cell to basement membrane. its mechanism was probably related to the inhibition of VEGF,adhesion molecule and MMP-2 expression, inducing endothelial and tumor cell apoptosis.
