Effect Of Coptis Injection On Febrility Nduced By Endotoxin In Rabbit And Its Mechanism

Coptis injection was prepared by mothod of extracted with water and precipitated with alcohol,its main component was berberine.It had displayed a wide range of pharmacological effect including clearing heat and dissipating, purging fire and removing toxic substance.It was used for relieving fever and resisting bacterium in clinically.The contemporary research indicated that coptis had many pharmacological action,such as antibiosis,antiendotoxin, antivirus,enhance immunization and relieve fever. In order to guide its application in clinic scientifically, the effect of coptis injection on febrility induced by endotoxin in rabbit and its mechanism were investigated by we.First, the active principle of coptis were extracted according the mothod of extracted with water and precipitated with alcohol to crank out coptis injection and it was maked quality standard research. It was yellowish-brown clear limpid sterile solution, its PH was 7.4.Second, the coptis injection depressant effect of Escherichia coli, Staphylococcus aureus and salmonella were determined through extraorgan bacteriostasis experiment. The consequence indicated that coptis injection had depressant effect of Escherichia coli, Staphylococcus aureus and salmonella, the diameter of inhibition zone was 2.15cm, 2.00cm, 2.57cm.The MIC was 0.25mg/ml, 0.25mg/ml,0.13mg/ml.Third, the febrile pattern of rabbits were insted according to escherichia coli endotoxin was taked into them. Coptis injection was used to treat them. In the process, body temperature of rabbits were surveyed uninterruptedly, the content of endotoxic in blood serum was determined, the number of white blood cell and classify of white blood cell were counted, the content of NO in blood serum,the activities of iNOS in blood serum and hypothalamus were all determined.The consequence indicated healthy rabbits temperature had risen after taken escherichia coli endotoxin into, their temperature had risen about 1℃after 1 hour and had obvious clinical febrile symptom, such as crouch,shiver,stick up hair,accelerated breathing,heart rate accelerating, but there were not eye conjunctival congestion. About 2 hours, the crouch and shiver had disappearanced, the coat were moistened,the respiration and respiration were more faster, body temperature of many rabbits were heightened obviously, external ears were vasodilatated and reddened,but there were not hemorrhagic erythema, there were consciousness, lassitude, pollakidipsia, the part had loose stools.The quantity of white blood cell were reduced at the early time and obviously increased in later period. The content of endotoxin and nitrogen monoxide were increased in the rabbit blood serum, the activities of iNOS in blood serum and hypothalamus were heighten. After injected the coptis injection, the rabbits body temperature were droped gradually, their clinical heat symptom had reduced, the content of white blood cell was obvious reduced in later period, the content of the endotoxin and the nitrogen monoxide (NO) were also obviously reduced in blood serum, the activities of the induction nitrogen monoxide synthase (iNOS) were also obviously reduced in the blood serum and the hypothalamus.It showed that body temperature of rabbits were positive correlation to the content of NO in the blood serum,the activities of iNOS in the blood serum and hypothalamus of rabbits.It explanated the rabbits were calefied by escherichia coli endotoxin,the calefied rabbits which causde by escherichia coli endotoxin were relieved fever by coptis injection, the content of NO in blood serum was degraded, the activities of iNOS in blood serum and hypothalamus and iNOS mRNA in hypothalamus were decreased. It prompted that the coptis injection cut down the content of NO according to depressant iNOS mRNA and restrain the activities of iNOS in hypothalamus,and then decrease the influence of NO contact with body temperature regulating center and achieved to degrade rabbits body temperature.
