Objective: This study was to investigate the distribution of polymorphism in the CAPN10 UCSNP43 G/A in population in Zhenjiang, and to investigate the genetic susceptibility of single nucleotide polymorphism (SNP) 43 in calpain-10 to type 2 diabetes (T2DM).Methods: A total of 225 people were involved in the study. Case-control study was conducted for 123 patients with T2DM (64 men and 59 women) and 102 normal control subjects (without T2DM family history, 55 men and 47 women). Mismatch polymerase chain reaction method was used to determine the distribution of allele and genotype frequencies of SNP43 in calpain-10. The frequencies of SNP-43 genotype and allele gene were compared between two subjects. Body mass index (BMI), waist-hip ratio (WHR), fasting plasma glucose (FPG), triglyceride (TG) and cholesterol (CHOL) were measured. Lastly the clinical dates were analyzed.Results: To the CAPN10 UCSNP43 polymorphism site, 1 with the homozygote mutation, 18 with the heterozygote mutation were found in 123 type 2 diabetes patients. 0 with the homozygote mutation, 23 with the heterozygote mutation were found in 102 healthy controls.â‘ In T2DM group, the frequency of G allele of SNP43 was 91.86%, the frequency of A allele was 8.14%. The distribution of allele was equal in two groups (P>0.05).â‘¡In T2DM group or in normal control group, the distribution of BMI, waist circumference, WHR, FPG, TG and CHOL of SNP43 GG was equal to those of SNP43 GA+AA(P>0.05).â‘¢In normal control group, the distribution of triglyceride (TG) of SNP43 GA+AA was higher than those of SNP43 GG (P<.05).Conclusion: There was UCSNP43 G/A polymorphism of calpain-10 gene in the Zhenjiang population, but the genetic susceptibility of SNP 43 in calpain-10 to T2DM could not be found in this study.
