| Objective:Along with the improvement of people's living standards, the change of habit in bite and sup and the trend of aged population, the incidence of hyperlipidemia disease have been higher and higher. It is closely related to the coronary heart disease and cerebrovascular disease. The increase of blood fat is prone to lead to atherosclerosis because lipids soakages make for aortic endarteria pathological changes, such as rigidity of blood vessel, lessening of inner pathway and fall of tension, which are easy to induce the obstacle of microcirculation and the shortage of tissues perfusion. Moreover, through the influence on double deck of lipid and the framework albumen, hyperlipidemia can increase the microviscosity and decrease the ability of distortion of erythrocyte. Due to the change of erythrocyte shape and the augmentation of erythrocyte membrane proportion, blood viscosity and hematocrit can be heightened, which can recede the microcirculation. As a result, blood and oxygen deficits of tissues lead to turbulence of metabolism. Usually, the cardio-cerebrovascular diseases present clinical symptoms after the changes of hemorheology. Consequently, it is significant to realize the hemorheological index of local healthy people, to research and to develop new style functional foodstuff and material medica, in order to prevent and cure cardio-cerebrovascular diseases. The experimentations proved that there had a negative correlation between the ingestion of polyunsaturated fatty acids (PUFA) in meals and the incidence of cardiovascular disease and the death rate. It is the mostly procreative area of zanthoxylum in Han Cheng, Shaanxi, China. The annual output of it preponderates over a quarter of a total countrywide. And the byproduct, the kernel of zanthoxylum, accounts for a greater part of total output of zanthoxylum. The content of PUFA in the kernel of zanthoxylum seed reaches to 90 percent. Thereinto, the content of required linoleic acid and linolenic acid in it reaches to 70 percent. Accordingly, it is very important to exploit the kernel of zanthoxylum seeds oil (KZSO). To investigate the preventive and therapeutical effects and mechanism of KZSO on hyperlipidemia, this experiment studied the hyperlipidemia rats to observe effects of KZSO.Methods:1. 3483 healthy cadres and employees of Xi'an who came to ambulatorium of our university to do physical examination, were excluded hepatic, nephritic and cardio-cerebrovascular diseases. Every examining index of them ranged in normal. They were divided into 5 groups according to the different ages both male and female, including 30-39group, 40-49group, 50-59group, 60-69group and over 70 group. We statistically analyzed their hemorheological indexes, such as high blood viscosity (HBV), low blood viscosity (LBV) and hematocrit (HCT), and compared those indexes with the references offered by apparatus in 2002.2. On the experiment, we chose thirty healthy SD rats. After one week adapted feeding, they were randomly divided into 3 groups, including control group, hyperlipidemia model group and KZSO group, each of 10, male and female half and half. Control group was fed by ordinary forage; model group was fed by lipid forage; KZSO group was garaged by KZSO besides lipid forage. Every group could take the forage and water at liberty. In addition, control group and model group were poured distilled water 2.5mL/kg/d, and KZSO group was poured KZSO 2.5mL/kg/d (equally ten times of adult dosage). The dosages were regulated in time along with the increase of weight. Blood samples were obtained by cutting necks after ten weeks feeding. The levels of blood lipids and the hemorheology parameters were determined; aortic tectological changes were observed by the pathological section with the HE dyeing; the activities of superoxidedismutase (SOD) and catalase(CAT), the content of malondialdehyde (MDA) and the restraining action to hydroxyl free radical in the liver tissues were measured.Results:1. It was showed by statistics of hemorheology that all the indexes of hemorheology in male were higher than those in female (P<0.01). The whole blood viscosity in female had an increasing trend along with age. However, the results of hemorheology in male showed that the indexes of 30-49 groups were higher than the other groups, and had a decreasing trend along with age over 50. The indexes of HBV, LBV and HCT in both male and female were higher than the references offered by apparatus in 2002.2. It was showed by zoopery that compared with the control group, cholesterol (CHOL), HBV, LBV were increased (P<0.05), and high density lipoprotein cholesterol (HDL-C) was decreased (P<0.05), which proved that hyperlipidemia model was built successfully. Compared with the model group, CHOL, triglyceride (TG), HBV of the group of KZSO were decreased (P<0.05); LBV was significantly decreased (P<0.01) and HDL-C was increased (P<0.05). There was no evident change in HCT. The activities of SOD and the restraining action to hydroxyl free radical in the liver tissues of KZSO group were significantly increased (P<0.01), while the activities of CAT were increased (P<0.05) and the content of MDA were decreased (P<0.05) compared with the model group. Aimed at the tectological changes, aortic endarteria of KZSO group were obviously slippery and its lipid's sediments were lightened distinctly compared with model group.Conclusion:1. The raise of blood viscosity of employees and cadres in Xi'an has a youthy trend. Clinical diagnosis towards hyperlipidemia should consider that such unpathological factors as age, sex, public structure and life habit affect the index of hemorheology."Healthy people"of this crowd, especially middle aged male, should regulate the bit and sup structures, improve the life habit, strengthen the physics exercises, and control their hemorheology level, in order to provide the foundations for the prevention and diagnosis of the cardio-cerebrovascular diseases. 2. As a kind of botanic oil containing rich PUFA, KZSO can modulate the blood lipid, improve the abnormal indexes in hemorheology, prevent lipid superoxidation, and protect endarterium. The mechanism may be related to its component, PUFA. |
