Aim: To study the relationship between CXCR4 and VEGF gene expression in osteosarcoma tissue, and to further compare CXCR4 and VEGF gene expression in isolated osteosarcoma patients with that in osteosarcoma complicated by lung metastasis patients.Methods: 45 isolated osteosarcoma patients and osteosarcoma complicated by lung metastasis patients were recruited in department of orthopaedics of the first affiliated hospital of Nanchang University. The immunohistochemical expression of chemokine CXCR4 and VEGF were analyzed using S-P immunohistochemical method.Results: VEGF expressed in cytoplasm of tumor cells and endothelial cells. CXCR4 expressed in nucleus, as well as in cytoplasm of tumor cells. The expression percentage of CXCR4 and VEGF gene in isolated osteosarcoma patients were 33 .3% and 59.3%, respectively. The expression percentage of CXCR4 and VEGF gene in osteosarcoma complicated by lung metastasis patients were 72.2% and 88.9%, respectively. The immunohistochemical expression of CXCR4 was correlated with the expression of VEGF gene in osteosarcoma tissue.Conclusion: The percentage of CXCR4 expression is significantly higher in osteosarcoma complicated by lung metastasis patients than isolated osteosarcoma patie -nts,and this chemokine can be considered as an important reference marker for osteosarcoma complicated by lung metastasis. There is not significant difference of percentage of VEGF expression between isolated osteosarcoma patients and osteosar -coma complicated by lung metastasis patients. However, there is significant correlation between the expression of CXCR4 and VEGF gene in osteosarcoma tissue.
