Objecitive1.To investigate the expression level of TS and the relationship with clinicopathologic parameters in rectal cancer。2.To investigate TS gene 5′-UTR polymorphism in rectal cancer and the relationship with clinicopathologic parameters。3.To analyse the relationship between the TS expression and TS gene 5′-UTR polymorphism in primary rectal cancer。MethodsExpression of TS in 40 primary rectal cancer tissues were analysed by means of immunohistochemistry,normal tissues of which were also assessed。PCR was used to detect TS 5′-UTR polymorphism in 40 rectal cancer tissues。Results1. TS high expression on tumor sections is higher than on normal tissues(P<0.05)。2.No significant association between the high expression of the TS and sex and age(P>0.05),but TS high expression was significantly relative to cell differentiation,infilatration,lymphatic metastasis and Dukes'stages(P<0.05)。3.PCR was used to detect TS 5′-UTR polymorphism in 40 rectal cancer tissues , only 32 rectal cancer tissues was successful。4. Type 2R/2R was 6.25%(2/32),type 2R/3R was 25%(8/32),type 3R/3R was 68.75%(22/32)。5. Over TS expression of type 3R/3R was 77.27%(17/22),which was higher than TS expression of type 2R/2R and 2R/3R, TS high expression of type 2R/2R and 2R/3R was 30%(3/10),the two groups have significant deviation(P<0.05)。6.TS gene 5′-UTR polymorphism was not correlated with sex,age,Dukes'stages,cell differentiation,infilatration,lymphatic metastasis(P>0.05)。Conclusion1.TS expression on tumor sections is higher than on normal rectal tissues。2.TS expression was significantly relative to cell differentiation,infilatration,lymphatic metastasis and Dukes'stages。 3.In primary rectal cancer ,type 3R/3R is the main part of TS gene 5′-UTR polymorphism。4.The TS expression of type 3R/3R was higher than type 2R/2R and 2R/3R。5.TS gene 5′-UTR polymorphism was not correlated with clinicopathologic parameters。...
