Objective To investigate the expression of RUNX3 and CyclinD1 of protein and mRNA in pancreatic carcinoma,and to analyze the relationship of them and clinical pathological parameters. Methods Expressions of RUNX3 and CyclinD1 in 47 cases of pancreatic carcinoma with surrounding normarl tissue, 18 cases with cystadenoma of pancreas and 12 nomal pancreas cases were detected by immunohistochemical SP method.Reverse transcription polymerase chain reaction (RT-PCR) was performed to detect the expressions of RUNX3 and CyclinD1 of mRNA in 42 cases of pancreatic carcinoma with surrounding normal tissue respectively.And their relationships with clinical pathological parameters were analyzed.Results (1) The positive expression of RUNX3 in pancreatic carcinoma was 57.45%(27/47), cystadenoma of pancreas 94.44%(17/18) and nomal pancreas cases 100%(12/12) (P<0.05). The positive expression of CyclinD1 in pancreatic carcinoma 72.34%(34/47) was significantly higher than that in cystadenoma of pancreas44.44%(8/18) and nomal pancreas cases8.33%(1/12) (P<0.05). The positive expression of RUNX3 in pancreatic carcinomas was 57.45%(27/47), which was significantly lower than those in surrounding normarl tissue. The positive expression of RUNX3 in surrounding normarl tissue was 85.11%(40/47).The difference of RUNX3 protein expression between two kinds of samples was statistically(χ2=8.782,P=0.003). The positive expression of CyclinD1 in pancreatic carcinomas was 72.34%(34/47), which was significantly higher than those in surrounding normarl tissue( 44.68%, 21/47)(χ2=7.406,P=0.007). (2) The relative expression value of RUNX3 mRNA found in 42 cases pancreatic carcinoma tissue was 0.2469±0.0708, which was significantly lower than those in surrounding normarl tissue( 0.7091±0.1764, t=15.7036,P< 0.001). The relative expression value of CyclinD1 mRNA found in in 42 cases pancreatic carcinoma tissue was 0.7769±0.1456, which was significantly higher than those in surrounding normarl tissue(0.2860±0.1224, t=18.9158,P<0.001). (3) the Expressions in pancreatic carcinoma were negatively correlated to histologic grade with clinical staging or node metastasis(P<0.05), however which were not in another clinical pathological parameters involving age and sex. (4) The expression of protein of RUNX3 was significantly correlated with Cyclin D1(r=-0.375, P=0.009), and the expression of mRNA of RUNX3 was correlated with Cyclin D1(r=-0.320, P=0.039). Conclusion (1) The differences of RUNX3 and CyclinD1 of protein expressions in pancreatic carcinoma, cystadenoma of pancreas and nomal pancreas were statistically significances. (2)The expressions of protein and mRNA of RUNX3 were weaker in pancreatic carcinoma than in surrounding normal tissues.The expressions of protein and mRNA of CyclinD1 were higher in pancreatic carcinoma than in surrounding normal tissues. The expressions of RUNX3 and CyclinD1 of protein and mRNA in pancreatic carcinoma were significantly correlated to histologic grade with node metastasis and clinical staging,which were not closely related to the age and sex. (3) The expressions of protein and mRNA of RUNX3 were weak in pancreatic carcinoma,and moreover, it was significantly correlated to histologic grade with node metastasis and clinical staging.The RUNX3 gene may be an anti-oncogene of pancreatic carcinoma. (4) The weak expressions of protein and mRNA of RUNX3 were correlated with CyclinD1 which were high expressions of them.when they act together ,they may play an important role in the carcinogenesis and progression of pancreatic carcinoma.
