Part I The expression of ICAM-1, P-selectin in the lung tissue of acute pulmonary embolism and the effects of Angelica injectionObjective: To discuss the expression of intercellular adhesion molecule-1, P-selectin in the vascular endothelial cell and the effects of angelica injection.Methods :Rats were randomly divided into control group(C-group), model group(M-group) and treatment group(T-group). Pulmonary thromboembolism(PTE) model was established by intravenous injection of auto-blood clots. The expression of ICAM-1, P-selectin protein were evaluated by immunohistochemical method and their mRNA contents were detected by in situ hybridization. Results : The notable embolism in pulmonary arteries, inflammatory cell infiltration around alveoli could be found by microscopy. The expression of ICAM-1 was elevated quickly at 3 hours after embolization (P<0.01).The expression of P-seletin was higher at 1 hour after embolization than that before embolization(P<0.01). Whereas, after using angelica injection, the expression of ICAM-1 and P-selectin were significantly decreased; the pathological injures was ameliorated.Conclusion : Angelica injection may alleviate pulmonary inflammation in rat pulmonary thromboembolism through inhibiting the expression of ICAM-1 and P-selectin.Part II The expression levels of cytokines in the lung tissue of acute pulmonary embolism and the effects of Angelica injectionObjective: To study the expression levels of endothelin-1(ET-1), cyclooxygenase-2(COX-2) in the lung tissue , the concentrations of thromboxane B2 (TXB2) and 6-keto- prostaglandin F1α(6-k-PGF1α) in the plasma in acute pulmonary thromboembolism(PTE), and to explore the effects of angelica injection on their expressions. Methods: Rats were randomly divuded into a blank group, a PTE model group,a angelica injection treat group.The PTE model was established by intravenous injection of autologous blood clots.The pathological changes of the lungs were examined with light microscope. By using immunohistochemistry, the expression levels of ET-1,COX-2 in lung tissue were examined.The plasma's concentrations of TXB2 and 6-k-PGF1αwere examined by radioimmunity assay.Result : Histopathological study showed severe lung injury in the PTE group,but mild in the angelica injection group. Immunohistochemistry study showed the difference between the blank group and the PTE group, the PTE group and the angelica group was significant (P<0.05).The rate of TXB2 and 6-k-PGF1α's concentrations (T/P)in the plasma study showed the difference between the blank group and the PTE group was significant (P<0.05). T/P difference between the blank group and the angelica group was slight(P>0.05).Conclusion :Angelica injection can reduce the expression levels of ET-1,COX-2 in acute PTE. Traditional Chinese medicine should be considered in acute PTE therapy.
