Objective To explore the influence and molecule mechanism of Angelica sinensis preparation and Sodium Ferulate to liver inflammation injury induced by lipopolysaccharide. Methods ICR mice were divided into five groups: Dex group .Angelica sinensis group ,Sodium Ferulate group, experimental control group, and normal group .To set up the model of Inflammation liver injury in administrating group and experimental control group which was induced by lipopolysaccharide in bacillus calmette-guerin primed mice. Angelica sinensis and Sodium Ferulate were administrated to the administrating group for ten days .One hour before we were ready to do the experiment,Dex was administrated to the Dex group.The same quantity brine was administrated to the experimental control group for ten days also. Observe and compare the liver inflammation injury of the mice through general pathology slice , and grade liver tissue pathology diagnoses; detecting the expression of TNF- α mRNA in the mice spleen monocyte through reverse transcription- polymerase chain reaction(RT-PCR). And then study the expression of TNF- α ,ICAM-1,E-selectin proteins in liver by SP immunohistochemistry. Results The experiments show that the liver inflammation injury of mice using Angelica sinensis ,Sodium Ferulate and Dex obviously lighter than the mice in the experimental control group (P<0.01) ; The expression of TNF- α mRNA in the spleen monocyte of liver inflammation injury mice increased obviously than the normal mice(P<0.01)at the 6th hour after injecting LPS .But the expression of TNF- α mRNA had no remarkable difference between the experiment control group and the administrating group. Study the expression of TNF- α protein in liver tissue, we found there are no obviously different between the groups using Angelica sinensis , Sodium Ferulate and the experimental control group ,and the expression of TNF- α protein in mice using Dex obviously lighter than the mice in the experimental control group (P<0.01); the expression of ICAM-l,E-selectin proteins in mice using drugs (Angelica sinensis , Sodium Ferulate and Dex)obviously lighter than the mice in the experimental control group (P<0.01) . Conclusions Angelica sinensis and Sodium Ferulate can restrain liver inflammation induced by lipopolysaccharide,and alleviate liver injury .but it maybe not work through influencing the expression of the TNF- α mRNA and pretion, but through controlling the expression of ICAM-1,E-selectin proteins, and then restrain that the inflammation cells adhere to VEC and soakage in liver tissue.
