End stage renal failure (ESRF) is all the renal diseases final results, the common pathological character is renal fiborsis. In this pathological process there are many primary and secondary metabolic disorders and syndromes,such as hyperlipea,hyperinsulinism,artherosclerosis,hypertension,Inflammatory reaction and etal. Hyperlipea has been noticed fully, it is thought to be the independent glomerular sclerosis risk factor. So through altering lipid metabolic disorders can postpone the procedure to ESRF.peroxisome proliferator-activated receptor-γ(PPAR-γ) is a member of nuclear hormone receptor super-family, and is highly expressed in kidney, adipose tissue. Activated by specific agonist, PPAR-γcan regulate target gene expression at transcription level. PPAR-γis involved in energy metabolism and lipocyte differentiation, Many scholars believe that PPAR-γhas a close connection with renal diseases and can be used as a new target site for the treatment of the urinary system disease. Transforming growth factor-β1 (TGF-β1) is the strongest factor inducing fibrosis, a lot of researches have recovered TGF-β1 have different effects: cell proliferation,cell differentiation,extracellular matrix cumulation,immunological function. Experiments within or without the living body have discovered PPAR-γcan inhibit the DAG-PKC-ERK singnal pathway and the activation of AP-I, and then to reduce TGF-β1. Another researches revealed that PPAR-γmay reduce the scavenger recepter-A (SR-A) of macrophage by 40%, which can intake LDL, especially ox-LDL. Mesangial cells and macrophage are homology, so Mesangial cells express SR-A, however if PPAR-γhas a effect on MC's SR-A still not be known.Abelmosehus is exstracted from sunset abelmoschus flower, pharmacology researches have discovered it has the effects such as antiinflammatory,lowering blood fat,clear oxygen free radica. The main element meletin can depress the proliferaton of MC and diminish ECM. Our object is to detect 5/6 nephrectomy rat's renal function,24 hour proteinurea,serum lipid,serium TGF-β1,pathological changes of the remain kidney, at last we analyize if they have connection with PPAR-γ,SR-A at gene leval.In our research we use 5/6 nephrectomy rat to set up ESRF model,at the twelth week,applying automatic biochemistry,ELISSA,RT-PCR and etc to detect above indexes. We drew the conclusions as follows: First,the 5/6nephrectomy rat can be used as animal model of chronic renal failure for experimental research。Second,when Abelmosehus reaches a certain concentration,it can reduce serum lipid and TGFβ1。Third,at above condition in the model group the PPAR-γgene expression in the renal cortex is in a lower level than in the sham group ,under the intervention of Abelmosehus PPAR-γincreased, there is statistical significance. Four,at the same condition the SA-Rgene expression in the renal cortex is in a hign level in the model group, under the intervention of Abelmosehus SR-A decreased, there is statistical significance. Five,Abelmosehus can protect remian renal function by lower serum lipid, at gene level it might through upregulating PPAR-γ,downregulating SR-A.。...
