Effects Of Acupoint Catgut Embedding On Lipid Metabolism And Liver PPAR Signaling Pathway In Nonalcoholic Fatty Liver Model Rats

The incidence of non-alcoholic fatty liver disease in humans and animals is increasing.It also increases the risk of type 2 diabetes,cardiovascular disease and chronic kidney disease.In this study,NAFLD model rats were induced by high fat diet,to explore the effect of single course of embedding acupuncture treatment on the lipid metabolism and liver PPAR signaling pathway.The rats were randomly divided into control group,control+embedding A group,control+embedding B group,model group,model+embedding A group,model+embedding B group and model+embedding C group.Collagen suture were implanted into embedding groups,Ganshu+Danshu+Houhai in A group and Ganshu+Danshu+Houhai+Housanli in B group,Zhouwan+Qihai+Tianshu in C group.After 2 weeks,the serum indicators and the contents of TG,TC,FFA,?-HB,ACC and FAS in the liver were measured.Observed the liver histomorphology of each group.The levels of mRNA and protein expression of liver AdipoR2,PPAR?(p-PPAR?)and CPT-1a were detected by RT-PCR and Western blot.The results revealed that:1.Model+embedding A group and model+embedding B group could increase the levels of serum HDL-C and ADPN in NAFLD model rats(P<0.05).On the opposite,they could reduce the levels of serum GLU,AST,AST/ALT,TG/HDL-C and TC/HDL-C in NAFLD model rats(P<0.05).2.Model+embedding A group could significantly reduce the TG content in the liver of NAFLD model rats(P<0.05).Model+embedding B group and model+embedding C group could cut down the contents of TG,FFA and FAS in the liver of NAFLD model rats obviously(P<0.05).3.Model+embedding A group,model+embedding B group and model+embedding C group could improved the degree of hepatic steatosis in NAFLD model rats.4.The expression of liver PPAR? mRNA and protein significantly increased in model+embedding A group and model+embedding B group,as well as the expression of liver p-PPAR? protein(P <0.05).Conclusions:1.Model+embedding A group and model+embedding B group could improve the blood lipid metabolisim and liver injury of NAFLD model rats.2.Both model+embedding A group and model+embedding B group could inhibit the lipid synthesis reaction in hepatocytes,which model+embedding B group was better than model+embedding A group.3.The effect in improved of model of liver lipid metabolism,which model+embedding A group and model+embedding B group did,may be involved in the increasing of the expression of liver PPAR? mRNA and protein.When it comes to clinical,the model+embedding B group could be a good choice.
