ObjectiveTo investigate the effect of the optic nerve injury to F-VEP(flash-visual evoked potential)and the change of CNTF(ciliary neurotrophic factor),CNTFR(ciliary neurotrophic factor receptor),BDNF(brain-derived neurotrophic factor)and OMgp(oligodendrocyte myelin glycoprotein)mRNA.Investigate the disposition of BDNF in the rats' retina and the optic nerve.And the effects of USW to the optic injury of rats.MethodsThis trial divide rats into three groups:the control group(only expose the optic nerve,not damage it),the injury group(expose and damage the optic nerve),the ultrashort wave therapy group.E stablish model of inadequate injury of rats'optic nerve.And then use the ultrashort wave cure the injury rats,which lasts 20d(7min every time,the output power is 40w,the frequency of waves is 43MHz).The changes of the optic nerve and retina were observed and the visual evoked potential response to flash(F-VEP)was recorded in normal,optic nerve injury and the USW therapy groups at the time of 1d,7d,14d,28d.Then analyse the change of rats'F-VEP.And use rt-PCR technique to measure the expression of CNTF CNTFR BDNF and OMgpmRNA at the time of 28 day after the injury.And investigate the disposition of BDNF in the rats' retina and the optic nerve.So we use F-VEP,immunohistochemical method and rt-PCR technique to evaluate the regeneration of optic nerves. ResultCompared with that of the normal rats,the latency of F-VEP in optic nerve crushed rats increased obviously after 2 weeks and the wave of F-VEP became lower and wider.And recovery response was observed after 2 weeks.The difference of the cure group's F-VEP and not cure group's F-VEP is not obvious(p>0.01).The injured expressions of CNTF,CNTFR,BDNF,OMgpmRNA were more than the normal (p<0.01).And mRNA of CNTF,CNTFR,BDNF,OMgp were expressed in normal REGs.Immunohistochemical results tell us that the expression of BDNF increase after injury and it is majorly distributed to inner nuclear layer and outer nuclear layer of retina.And rt-PCR point out that the cure group's BDNFmRNA were more than not cure group's.ConclusionsF-VEP correlate with the time of postinjury.14 days changed prominently and recovery start after 14 days.After the optic nerve injury,levels of CNTF CNTFR BDNF and OMgpmRNA increased.CNTF CNTFR BDNFmRNA are likely to protect the degenerarion of nerve cells by optic nerve injury,So exogenous CNTF might provide a supportive enviroment for axonal regeneration.The expression of BDNF increase after injury and it is majorly distributed to inner nuclear layer and outer nuclear layer of retina.USW could protect RGCs after the optic nerve injury.
