【Objective】Based on the good clinical effect of using electroacupuncture to treat the peripheral never, from the experimental aspect to explore the body movements and feeling functions'recovery of peripheral never injury after using the electroacupuncture, then through the studying of neurotrophic factor in the muscle to discuss its mechanism and approach.【Methods】Taking the SD rats to build sciatic nerve gripping damage model, choosing GB30, UB37, GB34, BL57, and positive pole on GB30, UB37 while negative pole on GB34, BL57,2mA electric current,2/100Hz frequency and 9min treatment. Then do the light pain threshold resistance and inclined plate experiment on the rats'both injury and normal posterior 1/3 of foot sole, from the behavior aspect to explore the body movements and feeling functions' recovery; through the calculation of gastrocnemius wet weight, the hematoxylin-eosin staining of gastrocnemius, sciatic nerve and spinal marrow, from the morphologic aspect to seek the evidence of sciatic nerve damage;Using western blot method for NGF,BDNF,CNTF,bFGF in the gastrocnemius, for gastrocnemius to reveal the possible mechanism of electroacupuncture treating the sciatic nerve damage, in order to further support using electroacupuncture to treat the peripheral never.【Results】1 EthologySolar-thermal pain threshold resistance:After 7 days, the sciatic nerve damage model group increase, compare with the normal group it has statistical significance; after 10 times electroacupuncture treatments, the electroacupuncture group is lower than model group, higher than normal group, and compare with the model group it has statistical significance. That means electroacupuncture can increase the sensitive of the pain, the sciatic nerve damage model is gradually recovering.Inclined plate test:After 7 days, the sciatic nerve damage model group's angle is lower than normal group, after 10 times electroacupuncture treatments, the electroacupuncture group's angle is higher than model group, control group, fake operation group, with statistical significance. It means the electroacupuncture can strengthen the rat's muscle power and improve the recovery of sciatic nerve.2 Pathologysciatic nerve:The normal and fake operation group's Schwann cell is marshalling, the model group's axon swelling, some are swallowed by macrophage, disorder ranged, glial cells obvious hyperplasia, the control group and model are similar, electroacupuncture group's axon and Schwann cell are normal.3 Neurotrophic FactorsGastrocnemius wet weight:After 7 days, the sciatic nerve damage model group is obvious decreased, compare with the normal group it has statistical significance; after 10 times electroacupuncture treatments, the electroacupuncture group and model group without statistical significance, but muscular recovery rate with statistical significance. It means the electroacupuncture treatment can promote the movements and muscular recovery.Gastrocnemius western blot:After 7 days damage of the sciatic nerve, the NGF,CNTF,BDNF,bFGF is highest in model group, compare with the normal group it has statistical significance; after 10 times electroacupuncture treatments, the electroacupuncture group are lower and with statistical significance. It means the NGF,CNTF,BDNF,bFGF keep increasing after damage of the sciatic nerve and electroacupuncture therapy can let them down which reveal that under the damage of the sciatic nerve condition, the target muscles loose the innervations, the body would secrete more these kind of factors to protect, and after treatment, the nerve is recovering, these kind of factors become less.Spinal cord immunohistochemical:the light density of NGF,CNTF,BDNF,bFGF in electroacupuncture group is higher than control and normal group, with statistical significance; it means the electroacupuncture method can enhance them, and body release more after the sciatic nerve damage to protect the spinal cord neurons.【Conclusion】The research is taking the solar-thermal pain threshold resistance and inclined plate test as index, observe the feeling and movements condition after electroacupuncture treatment. The result shows that it can increase the sensitivity of pain and muscle recovery rate, the possible mechanism is the repairmen of sciatic nerve, the foundation of pathway and protection of spinal cord neurons.After observation of hematoxylin-eosin staining on sciatic nerve and medulla, it shows that the electroacupuncture treatment can protect sciatic nerve and spinal cord neurons, promote repairmen of sciatic nerve.The western blot detection of NGF,CNTF,BDNF,bFGF in gastrocnemius and immunohistochemistry detection of NGF,CNTF,BDNF,bFGF in spinal cord neurons shows that electroacupuncture treatment can increase the neurotrophic factor. The possible mechanism is with the repairmen of sciatic nerve, the axoplasm can retrogradation, the neurotrophic factors are transported to protect spinal cord neurons. Taking all the results as a whole, we can primarily deduce that after electroacupuncture treatment the sciatic nerve injury and feeling, movements function are enhanced, the mechanism is the repair of the nerves, let the NGF,CNTF,BDNF,bFGF in the target muscle through the axoplasm to retrograde in order to protect spinal cord neurons.
