ObjectiveTo investigate the significance of heparananse(HPA)and its correlation with asic fibroblas growth factor(bFGF)in cervival cancer by detecting the expression of HPA and bFGF in cervical intraepithelial neoplasiaⅢ(CINⅢ)and cervival cancer, and analyzing the association between the two factors.MethodsImmunohistochemical methods were used to detect the protein expression of HPA and bFGF in paraffin-embedded specimens of 88 cervical cancer,21CINⅢand 20 normal cervical tissue. The correlation of the two factors with the clinical features of cervical cancer was evaluated by chi-square test, and the relationship of the two factors was analyzde by Spearman rank correlation test.Results1.The positive percentage of expressions of HPA in normal cervical tissue,CINⅢand cervical cancer were 5.0%,14.3%and 63.6%,respectively.The positive percent of HPA in CINⅢwas not significantly higher than normal cervical tissues(p=0.317),and which in cervical cancer was significantly higher than CINⅢand normal cervical tissues(p=0.000).2.The positive percentage of expressions of HPA in different stage cervical cancer was stageⅠ44.1%,stageⅡ75.0%and stageⅢ,Ⅳ76.9%.There was significantly difference between stageⅠand stageⅡ(p=0.014),stageⅠand stageⅢ,Ⅳ(p =0.011),but no significantly difference between stageⅡand stageⅢ,Ⅳ(p=0.011). The positive percentage of expressions of HPA in lymphatic metastasis positive and negative groups were 100%and 8.6%,and there was significantly difference between them(p=0.000).The positive percentage of expressions of HPA in<40 years group and≥40 years group ge were 67.7%and 61.4%,and there was no significantly difference between them(p=0.555).The positive percentage of expressions of HPA in different groups of squamous cell carcinoma and adenocarcinoma were 63.3%and 66.7%,and there was no significantly difference between them(p=0.092).The positive percentage of expressions of HPA in high, middle and low histological grading cervical cancer were 63.2%,62.2%and 66.7%,and there was no significantly difference amongthem(p=0.934).3.The positive percentage of expressions of bFGF in normal cervical tissue, CINⅢand cervical cancer were 10.0%,28.6%and67.0%,respectively.The positive percent of bFGF in CINⅢwas not significantly higher than normal cervical tissues(p>0.05),which in cervical cancer was significantly higher than CINⅢand normal cervical tissues(p<0.05).4.The positive percentage of expressions of bFGF in different stage cervical cancer was stageⅠ47.1%,stageⅡ78.6%and stageⅢ,Ⅳ73.7%.There was significantly difference between stageⅠand stageⅡ(p=0.011),stageⅠand stageⅢ,Ⅳ(p=0.043),but no significantly difference between stageⅡand stageⅢ,Ⅳ(p =0.637).The positive percentage of expressions of bFGF in lymphatic metastasis positive and negative groups were 100%and 32.1%,and there was significantly difference between them(p=0.000).The positive percentage of expressions of bFGF in<40 years group and≥40 years group ge were 61.3%and 66.7%,and there was no significantly difference between them(p=0.614).The positive percentage of expressions of bFGF in different groups of squamous cell carcinoma and adenocarcinoma were 64.6%and 66.7%,and there was no significantly difference between them(p=0.980).The positive percentage of expressions of bFGF in high, middle and low histological grading cervical cancer were 68.4%,60.0%and 70.8%, and there was no significantly difference between them(p=0.632). 5.The expression of HPA had positive correlation with the bFGF by Spearman rank correlation test(r=0.530,p=0.000).Conclusion1.The positive expression rate of HPA and bFGF in cervical cancer was significantly higher than CINⅢand normal cervical tissue, suggested that HPA and bFGF possibly played an important role in the genesis and development of cervical cancer.2.The expressions of HPA and bFGF had significant relation with clinical stage and lymphatic metastasis but they had no relation with the histological classification, histological grading and age.So they may be the important referenced indexs of predicting biological behavior and valuable targets of the treatment of cervical cancer.3.The expression of HPA had positive correlation with bFGF in cervical cancer, suggested that HPA and bFGF possibly played an important role in the genesis, development, invasion and metastasis of cervical cancer together.
