ObjectiveTo investigate the effect and the mechanism of lumbrokinase on the skeletal muscles in the ischemic hindlimbs of rats at different times.MethodOne hundred and fifty adult Wistar rats were distributed to lumbrokinase treating group, non-treating group and control group randomly, after finishing the ischemic hindlimb model by microsurgery technique. The rats were sacrificed at 3, 7, 14, 21and 28 days after the operation respectively. The extensor digitorum longus, gastroenemius and soleus muscles in the ischemia hindlimbs and the opposite normal hindlimbs were harvested for the further research. The water content, free calcium concentration of the mitochondrial, the opening level of mitochondria permeability transition pore (MPTP) in the ischemic skeletal muscles from the 3 groups was detected respectively.ResultsThe water content of extensor digitorum longus in the treating group was higher than that in the non-treating group from 14 days after surgery(P<0.05); And the water content of gastroenemius and soleus muscles in the treating group was higher than that in the non-treating group from 21days (P<0.05) . The free calcium concentration of extensor digitorum longus was lower in the treating group than that in the non-treating group at 7, 14 days after surgery (P<0.05) ; The free calcium concentration of gastroenemius was lower in the treating group at 14, 21 days after surgery (P<0.05) ; And the free calcium concentration of soleus muscle in the treating group was higher at 7,14, 21 days (P<0.05) . At 28 days after operation, the free calcium concentration of the mitochondrial has no significant difference between the treating group and the non-treating group in these three skeletal muscles. The MPTP opening level (expressed by AS) was higher in the treating group than that in the non-treating group at 14, 21, 28 days after surgery in these three skeletal muscles.ConclusionLumbrokinase could decline the loss of water content in the later period of ischemia, decrease the concentration of free calcium in the mitochondrial and inhibit the opening level of the MPTP in the ischemic skeletal muscles of rats, which suggested that lumbrokinase was effective on treating the ischemic hindlimb muscles of rats.
