Studies have shown that chronic inflammation plays an important role in benign prostatic hyperplasia (BPH) development. The experiment based on the fact that n-3 polyunsaturated fatty acid can change the content of inflammation cytokines and eases the benign prostatic hyperplasia inflammation. This research also provide the possibility for us to chose new treatment medicine.However, experimental study showed that large intake of n-3 polyunsaturated fatty acids have a negative impact on the body so that we should consider the ratio of n-6/n-3.This experiment established model of benign prostatic hyperplasia in mice. We adjusted the ratio of n-6/n-3 and observated the ratio of n-6/n-3=5group which may have an effect on benign prostatic hyperplasia. Mice were randomly divided into three groups: normal control group, the ratio of n-6/n-3=5group, the control model of benign prostatic hyperplasia group. Four weeks later we executed the mouse and detected spleen index and prostate index together. The ratio of n-6/n-3=5 group showed that epithelial monolayer partly papillary hyperplasia prominent to the prostate cavity. Only a small amount of pink secretions was observed in the ratio of n-6/n-3=5group when compared with the normal group.The experimental results show that the ratio of n-6/n-3=5 group to a certain extent have an effect on benign prostatic hyperplasia model mice and reduce the mice spleen index and prostate index.In order to declear the ratio of n-6/n-3 which may be more effective we did the experiment followed.Mice were randomly divided into six groups: normal control group, the control model of benign prostatic hyperplasia group, the ratio of n-6/n-3=1.7group, the ratio of n-6/n-3=5group, the ratio of n-6/n-3=10group, finasteride treatment group. Besides the normal control groups all groups were injected testosterone propionate 5mg/kg·d subcutaneously according to the weight with regulation. Executed mouse after six weeks of experiment and detected the index of the spleen and prostate index.We had a detection with IL-1β, TNF-αand IL-6 levels as well as the content of DHT in blood serum. We had a detection of the levels of prostate acid phosphatase in the Prostate tissue.The result of this experiment showed that the ratio of n-6/n-3=1.7 group and the ratio of n-6/n-3=5 group can significantly reduce benign prostatic hyperplasia index (P<0.01) and prostatic acid phosphatase (P<0.01). This group significantly inhibited prostate hyperplasia and growth situation was obviously better than the model group.Experiments showed that the ratio of n-6/n-3=1.7 group and the ratio of n-6/n-3=5 group had an declining trend about the proinflammatory cytokines, IL-1β, TNF-α, IL-6 content and showed inhibition on the prostate hyperplasia mice .We could see the biggest drop in the ratio of n-6/n-3=5 group. It shows that the sources of CO2 supercritical extraction from rape pollen have an effect on benign prostatic hyperplasia by regulate the ratio of n-6/n-3 and have a certain role in immune regulation. The experiment results showed that dietary adjustments in the ratio of n-6/n-3=5 group and the ratio of n-6/n-3=1.7group had a certain role in immune regulation and anti-inflammatory effects on benign prostatic hyperplasia mice. The fact also showed the ratio of n-6/n-3=5 is more effective. The ratio of n-6/n-3 =5 group about prostatic acid phosphatase in the lower level are better than the ratio of n-6/n-3=1.7 group. The ratio of n-6/n-3=5 groups show little prostate cavity pink acid secretion. We speculate the mechanism of inhibiting prostatic hyperplasia may be specific to the male hormone and binded to dihydrotestosterone with prostate androgen receptor in order to prevent the receptor play a role as a transcription factor. However the ratio of n-6/n-3=1.7 had significantly lower dihydrotestosterone content in the serum. We speculate that with the ratio of n-6/n-3 increased it may have effect on steroid metabolism. The specific mechanism need to be further explored.
