Objective To investigate the serum lipid levels features of the patients with Alzheimer's disease (AD) of the Xinjiang Uygur and Han nationalities , and the impact of APOE gene polymorphism on lipid metabolism and the role in pathogenesis of AD. Methods From Oct.2004 to May 2007,an AD epidemiological investigation was carried out among the permanent residents(age≥50),including both Uygur and Han nationalities in Xinjiang. Polymerase chain reaction - restriction fragment length polymorphism analysis (PCR-RFLP) is practiced on potential 157 cases (65 cases Han,92cases Uygus)of AD and 155 cases(68cases Han,87cases Uygus) of control to analysis all Apoe genotypes and allele frequency, and measure the concentration of plasma cholesterol (TC),and triglyceride (TG).Result (1) Mean of serum TC AD is 4.81±1.19mmol / L, serum TC control group mean is 4.45±1.05 mmol / L, and TC levels is statistically difference between AD and control(t =2.81, P <0.05). AD serum TG median is 1.60mmol/L, the contrast TG median is 1.37mmol/L, and TG levels is statistically difference between AD and control (p<0.05). TC and TG mean in AD group are higher than in the contrast group; AD patients's serum TC of Uygur mean is 4.75±1.35mmol/L,TG median is 1.75 mmol/L. AD patients's serum TC of Han mean is 4.90±0.91mmol/L,TG median is 1.47 mmol/L. Differences between the two groups is of no statistical meaning(P >0.05) . (2)The average number of AD group mean TC in Apoeε4 group is 5.17±1.12mmol/L while the control group is 4.52+0.77mnol/L, which represent statistic difference between their TC level(t=2.62, P <0.05 ). The average number of Apoeε4 group mean TC in group AD is 5.17+1.12mmol/L , Apoeε3 group mean TC in group AD is 4.66+1.21mmol/L , Apoeε2 group mean TC in group AD is 4.55+1.11mmol/L , among the three groups exist statistical difference (F=3.65, P <0.05).In the three Apoe groups different ethnic of Uygur and Han nationality in AD see no difference in the levels of the lipids(P>0.05).The relationship between ApoE phenotype and lipids was similar in the two nationalities. (3)The non-conditional logistic regression analysis with the limitation of age and sex showed that the ApoEε4 allele (OR=2.28, 95%CI=0.895-1.305,P=0.004)and TC is the risk factor of sporadic AD. Conclusion Our data suggested that AD patients carried dyslipidemia. TC and TG mean in AD group is higher than in the contrast group. ApoE genotypes probably regulated lipid metabolism. Apoeε4 allele and TC might be a risk factor in Sporadic AD.
