Objective:The uterine cervix cancer is one of the most common malignant tumors for females.It is the second malignant tumor for world-wide females and the first for Chinese females.Its high death rate threatens the women's health acutely.Statistics shows that about 500,000 women are diagnosed as uterine cervix cancer,and about 250,000 dies of it every year.The uterine cervix cancer is also very common in Xinjiang Vygur Autonomous Region,and doctors are always focusing much attention on it. Infected HPVs,such as HPV 16 and HPV18,are the main cause of uterine cervix cancer, while the protein abnormities are induced by gene expressions in cells and thus the cancer is.caused by a virus.So determining the related genes and understanding the mechanism of cell cancerization can provide molecule symbols and target genes for clinical diagnosis,prevention,prediction and therapy of the uterine cervix cancer.To understand the changes in gene expressions of patients using Oligo chips,find the rules and internal correlations between differentially expressed genes and its bionomics. Methods:7 samples of cervix prickle cell carcinoma and 4 normal samples are collected,from Uygur female patients in our hospital from December 2006 to May 2007, who come from the southern Xinjiang.The total RNA from samples are extract by Trizol One-step Method.While cRNA probes ere being reversely transcript synthesized, 11 samples and Universal Hunman Reference RNA samples are marked by Cy3 and Cy5 respectively.After the Cy3-marked probes with the Cy5-marked probes have been mixed, they are hybridized with the human expression chips containing 21522 genes.When chips are laundered,scanned and processed using software,the differences between the uterine cervix cancer expression profiling and normal sample expression profiling are analyzed,and the differentially expressed genes are analyzed using cluster analysis methods.Results:Compared with normal gene group,the uterine cervix cancer group has 1159 differentially expressed genes which changed more than 2 times and 378 differentially expressed genes which changed more than 3 times.In this paper 212 differentially expressed genes are mainly discussed,which significant deviation is bigger than 4.And the 212 genes include 166 upregulated genes and 46 downregulated genes. The results of cluster analyses show that individuals in a cluster have same characteristics;individuals in different clusters have significantly different characteristics. Conclusions:Very few differentially expressed genes from female Uygur patients have been found.Based on this,the relations between variations of gene difference changes and biological behavior can be predicted to some degree.
