Objective: @①By detecting the 6 antibodies which have appeared before HCC in patients with chronic Hepatitis B,we evaluated the clinical significance of these 6 antibodies, and the clinical diagnosis value of different assembly of them.②To study the effect of TCM in preventing the development of HCC from HBV-associated cirrhosis by observing the efficacy of compound phyllanthus urinaria L on 52 patients with HBV-associated cirrhosis who may develop HCC. @Methods: @X-gene positive and negative HepG2 cells.were synthesized and subjected to cDNA subtraction.Characterization of several differentially expressed genes have shown that they enhance hepatocellular growth in culture and in soft agar.Specific ELISAs, sequences spanning hydrophilic domains were selected for solid-phase peptide synthesis, the peptide used for coating well. These antibodies include anti-URG4,anti-URG7,anti-URG11,anti-URG12,anti-URG19 and anti-DRG2.830 serum samples were collected from Chinese and Korean.Among them,416 were HBsAg[-]and 398 were HBV carriers with chronic liver disease.102 patients of liver cirrhosis with regen-nodule due to hepatitis B were randomly divided into Phyllanthus urinaria L group(n=52) and control group(n=50).To observe the clinical symptoms, biochemical indicator, levels of HBV,3 indexes of liver fibrosis, and the antibodies distribution change and the risk of developing HCC. @Result: @①Antibodies was statistical differently compared with HBsAg[-]in patients with HBV-associated cirrhosis and HCC(P=0.0000),but no statistical differently in most carriers without active liver disease (P=0.1278),and in sera from uninfected individuals, with tumors other than HCC(P=0.3462~0.7216)and those with non viral hepatitis(P= 0.4548).At high risk of developing HCC,these antibodies were found to be independent of nationality and ethnicity.②After treatment there are statistical significance during the control group and treatment group in the clinical symptoms, biochemical indicator and 3 indexes of liver fibrosis(P<0.01or(P<005)),two groups have no statistical different on the levels of HBV,Among 102 patients,2 patients of treatment group developed into HCC,while the number of control group was 9,and the disparation between 2 groups showed statistical significance(P=0.0212). Contrasted to the control group, the number of antibodies of treatment group was distinctly decreased after 24mo(P<0.001).Anti-URG11 and anti-URG12 had the highest positive frequency, all above 60%in 2 groups. And the number of these two antibodies of treatment group was both decreased after treatment, while in control group was rose, and the disparation between two groups showed statistical significance(P=0.0417,P=0.0452).Follow-up study of 11 patients who developed into HCC had confired this result. @Oonclusion: @①These antibodies be detected to find patient with Hepatitis B Virus-Associated showed differently positive rate of these antibodies, and these antibodies can be detected before the appearance of HCC for several months or years, which suggest that they may be served as preneoplastic markers for patientswith HBV and have prognostic value in predicting the outcome of HBV-associated cirrhosis who may develop HCC. and may be identified by a simple blood test.②Anti-URG11,anti-URG12 and DRG2 may be served as indexes to predict the outcome and therapeutic efficacy of HBV-associated cirrhosis who may develop HCC.Through the treatment of compound phyllanthus urinaria L,we found that traditional Chinese medicine may prevent the development of HCC from HBV-associated cirrhosis to some extent. The clinical assessmen of this study should take more samples and much more time to research, and a longtime treatment of traditional Chinese medicine was the key point.
