Background:The loss of cadiomyocytes and reconstitutione of cardiac ventricle after Acute myocardial infarction(AMI)is the main cause inducing heart failure and clinical death.Recently,because of the treatment methods (including intervention,operation and drug treatment),The infarct-related artery can be reperfused effectively.But myocardial infarction has usually generated quickly before the opening of the infarction artery.So the infarction of the myocardial function can not be improved by these treatments.The development of stem cell technology brings the hope to the treatment of acute myocardial infarction and becomes the hotspot of the current study.Bone marrow stem cell is one type of the adult stem cells being studied most extensively.It is a group of stem cells which has multi-directional differentiation potentiality and exits in cavitas medullaris,having the ability of differentiation to myocardial cell,vascular endothelial cell and smooth muscle cell.Bone marrow stem cells have many merits like easy gaining,hypotoxicity or avirulent,once transplantation utility in long term,no immune rejection in autologous transplantation and so on. Besides,due to the fact that the bone marrow stem cells can be taken from the patient himself,there is no controversy on ethics.These features make bone marrow stem cell a very important source of the regenerative medicine research.Objective:Discuss the effect of the transplantation of the rabbit's autologous bone marrow stem cell(mononuclear bone marrow cells,mesenchymal stem cells)on the treatment of acute myocardial infarction(AMI) and the survival of bone marrow-derived mesenchymal stem cells in the myocardial infarction zone.To investigate the effect of autologous bone-marrow stem cells (mononuclear bone marrow cells,mesenchymal stem cells)were transplanted into the acute myocardial infartion.Methods:Models of AMI are established by ligation of left anterior descending artery in rabbits.Bone marrow is aspirated from the femoral bone of the rabbits in the condition of asepsis.The MBMCs and MSCs are separated, purified and cultivated.Twenty-four survived rabbits are randomly divided into control group(groupⅠ,n= 8),MSCs group(groupⅡ,n=8,MBMCs group (groupⅢ,n=8.The DMEM are injected into scar of myocardium in groupⅠ. The MSCs labeled by 5-Brdu are injected into groupⅡand groupⅢwith MBMCs._Specimens are taken 4 weeks after the implantation to measure the heart function of each group by echocardiography and to detect the differentiation of stem cell by using immunity histochemistry.Result:Echocardiography indicates that:Compared with control group,4 weeks after the LVEF and LVFS of MBMCs group and MSCs group improved significantly(P<0.05).Pathology indicats that brdu positive cells were survived,cultivanted and differentiated into cardiomyocytes in MSCs group.conclusion MSCs can survive,and differentiate into cardiomyocytes, improve myocardial perfusion and cardiac function when transplanted into ischemic myocardium.
