| Candida albicans is a kind of normal parasitic microflora on the skin and mucocutaneous membrane of healthy individuals. However, when immune defenses are compromised or the normal microflora balance is disrupted, C. albicans can be transformed by itself into an opportunistic pathogenic killer. Recently, the interventional therapy such as the frequent use of immunosuppressants and chemotherapeutic drugs for cancers has increased the susceptivity and morbility of patients. So far, the methods used in clinic last several days, and the sensitivity is very low. So, it is significant to develop a new way for diagnoses.Heat shock protein 90 (HSP90) is a virulent factor of C.albicans. And HSP90 is a dominant antigen of C.albicans. It can induce the production of special antibody. The studies of Matthews and our Lab have demonstrated that the antibody is protective.We have constructed the hybrid phage displaying the H epitope of HSP90, and stored the recombinate plasmid.We prepared the hybrid phage displaying the H epitope of HSP90 and determined its immunity. Then, the hybrid phage was used as coating antigen for establishing the ELISA, and the serum of seven hundred and six patients with cancer was detected. The results were as follows:1. The hybrid phage displaying the H epitope of HSP90 can react with special antibody in serum of patient with candidiasis. So, the hybrid phage can be used to detect candidiasis.2. We determined the concentration of coating antigen is 70μg/ml and the serum should be diluted 1/200. The cut-off value is 1.5.3. We detected the special antibody in thirty one of even hundred and six patients with cancer. Two of thirty one patients have been demonstrated as candidiasis by clinical. We also demonstrated the existence of antibody by Western bolt. The ELISA was so effective that it may be a new way for diagnosis of C.albicans infection. |
