C.albicans is a kind of opportunistic pathogen on normal organism. However, when immune defenses are compromised or the normal microflora balance is disrupted, C.albicans can be transformed by itself into a killer.The secretion aspartic acid proteinase 2(Sap2) is a virulent factor secreted by C.albicans. Many experiments demonstrate that Sap2, as a immune superior antigen, shows remarkable immune protective function to animals from sublethal disseminated candidiasis. The anti-Sap2 immune body also has obvious function in the process of passive immune treatment to animals infected by C.albicans.As a new multi-peptides carrier, phage plays a very important role in immunology. The two kind of epitopes of VKYTS and SLAQVKYTSASS of Sap2 have been displayed on the surface of phage respectively. This article studied the protective function against candidiasis in mice with the two kind of hybrid phages Moreover, The passive immune treatment function in mice infected by C.albicans with the antibodies to the two kind of hybrid phages was also be studied. BALB/c mice were immunized intraperitoneally with Sap2 recombinant protein,hybrid phage displaying epitope of VKYTS,hybrid phage displaying epitope ofSLAQVKYTSASS and wild-type phage respectively. The high titre antibody was obtained from the immunized mouse blood serum. This demonstrate that phage, As a multi-peptides carrier, is possible to induce organism to produce the intense immune response without any additional adjuvant. The mice after the fourth immunity were infected with ATCC10231 in the safe concentration(2×106cells)by intravenous inoculation, then the pathology changes in liver and kidney were observed. It is discovered that all of the two kind of hybrid phage and the Sap2 recombinant protein showed certain immune protective function to animals against candidiasis. The mice infected with ATCC10231 in the death concentration (2×107cells)were passive immune treated with the four kind of Multi-clone antibody produced above, all of the antibodies to the two kind of hybrid phage and the Sap2 recombinant protein showed certain passive immune treatment function in mice infected with Candida albicans.
