Objective To investigate the changes of motor and sensory function recovery,as well as proteomics expression changes of lesioned spinal cord and associated tissues after complete transected spinal cord(cTSC)subjected to Dumai electric acupuncture stimulation(EA)in rats.Methods The 56 femal and SD rats were randomly divided into sham-operation control group (SO),the 3rd,14thand 28thday group after cTSC or EA,each group has 8 animals.All experimental animals were performed the complete transected spinal cord at T11 vertebral body level except that the animals of SO group were only performed laminectomy.The acupoint of EA was divided into two group containing Dazhui,Mingmen and Zhongshu,Xuanshu,every group were stimulated in turn every other day.The cortical somatosensory evoked potential(CSEP)was detected in the SO group and all pre-operated animals were performed BBB scoring,and the BBB scoring was assessed again at 1st,3rd,70th,140th,21stand 28thday after cTSC and EA,and then the CSEP was registed again at 14thand 28thday after cTSC and EA. The animals of SO group were permited survival 3 days,and the others animals were executed the 3rd,14thand 28thday after cTSC and EA.The tissues samples were extracted from the cerebral cortex,the caudal and rostral of lesioned spinal cord,and denervated muscle in rats at the corresponding times period after cTSC and EA,only the scar tissues of spinal cord were obtained at last two times period.The differentially expressed proteins were seperated initially by 2-dimensional electrophoresis and identified with Mass spectroscopy among different groups.Results Except the 1stand 3rdday,the BBB scoring displayed significant increasing in the EA group,comared with the cTSC group,but their BBB scoring still lowed obviously the SO group. Contrast to the CSEP,the P1-N1 wave of CSEP was only registed at 14thand 28thday after EA, and the P1-N1 amplitude has not statistic significance at the 14thday after EA,compared with SO group,following the P1 and N1 latency took on primitively the same level at 28thday after EA compared with SO group.There were 64 kinds of proteins differentially expressed among these groups,their expression showed up-and down-regulated or losed changes in the tissues after EA.And these differential expressed proteins were divided different category into the different function,which including agnoprotein,enzyme-protein,cytoskeletal protein,heat shock protein,antioxyge protein,signal transduce protein,structural protein,molecular chaperones and energy metabolism enzyme proteit.Conclusion The study deemed EA could partly promote the recovery of motor and sensory function after cTSC.There also displayed the EA could stimulate some proteins expression of the corresponding tissures after cTSC,which participated in promoting the survival of injuryed neurons and their axis regeneration by up-and down-regulating or losing expression after cTSC.
