Regulatory T cells is one kinds of immunological regulatory cells, it mainly endures by the driving way induction and the maintenance organism circumference immunity, the adjustment organism immunologic balance, many research discovered it, in prevents and suppresses in own immunological disease, the inflammation, allergy and transplant rejection in a very important role.Chronic renal failure can be the development of kidney damage with the increased gradually and is divided into the following four stages: decompensation, discompensation period, and the failure of uremia period. Chronic renal insufficiency often with immune dysfunction, and abnormal immune cells to the main.Regulatory T cells play an important role in maintaining immunologic homeostasis and preventing autoimmunity. The peripheral blood samples were collected from 50 patients with chronic renal insufficient to investigate the significance of CD4+CD127-regulatory T cells in peripheral blood of patients with uremia and renal allograft recipients. The CD4+Tcell ratios in lymphocyte, CD4+CD127-Treg and CD4+CD25+CD127-Treg ratios in CD4+T were detected by using flow cytometry. We found that the CD4+T ratios in lymphocyte of chronic renal insufficient were higher than healthy control group and there wasn't significantly difference between the CD4+CD25+CD127-Treg ratios in CD4+T of chronic renal insufficient and that of healthy central. The CD4+T ratios in lymphocyte of chronic renal insufficient were lower than healthy control group except compensatory phase. And there was no correlation between CD4+Tcell ratios in lymphocyte, CD4+CD127-Treg, CD4+CD25+CD127-Treg ratios in CD4+T and the value of BUN, Cr among the hypertension patients. The number of CD4+T is increasing, CD4+CD127-Treg is decreasing in the patients with chronic renal insufficient and their immune is disturbing.The peripheral blood samples were collected from 13 maintenance hemodialysis patients,before trans-analysis and after. The CD4+CD127-Treg and CD4+CD25+CD127-Treg ratios in CD4+T were detected by using flow cytometry to investigate the significance of CD4+CD127- regulatory T cells in peripheral blood of maintenance hemodialysis patients. It can investigate that the CD4+T ratios in lymphocyte of maintenance hemodialysis patients were higher than healthy controls group before trans-analysis and the CD4+CD127-Treg and CD4+CD25+CD127-Treg ratios in CD4+T were lower, but CD4+T ratios decreased and CD4+CD25+CD127-Treg ratios increased after trans-analysis. The number of CD4+T is increasing, CD4+CD127-Treg is decreasing in maintenance hemodialysis patients and their immune is disturbing, and hemodialysis can improve immune function of the patients.Kidney transplantation is ideal uremic patients, the economy, and may significantly improve the quality of life of a treatment methods for uremia patient at present, but rejection and the long-term use of immunosuppressive drugs side effects, including immune suppression causing by infection, the increasing incidence of cancer and other serious affected graft and the survival.How to control organ transplant rejection, graft induced by the host immune tolerance has been research in the field of transplantation immunology the hot spot and difficult problem. In vivo and vitro through antigen-specific or non-specific amplification of the effect of reduced immune cell function and weaken their proliferative capacity, to suppress or control the level of immune response.Renal transplant recipients were divided into chronic rejection group and stable group, In this experiment, apply to monoclonal antibody flow cytometry detect the level of CD4+CD127-regulatory T cells in peripheral blood of patients with uremia and renal allograft recipients, and then compared to normal control group, to investigate the significance of CD4+CD127-Treg,CD4+CD25+CD127-Treg in peripheral blood with the healthy control group for comparison, to investigate the significance of the heterologous organ to transplant tolerance and overcome the rejection reaction to provide some references. We found that the CD4+T ratios in lymphocyte of chronic rejection group were higher than healthy controls group and stable group, the CD4+CD127-Treg ratios in CD4+T of chronic rejection group were lower than normal control group and stable group, the CD4+CD25+CD127-Treg ratios of renal chreonic rejection group were lower than normal control group. It gave us inspiration that if promotes the Treg g development and proliferation, will be able to effectively enhance the immune tolerance and reduce the amount of immunosuppressive drugs.
