The Effects Of TAP On Diabetic Nephropathy And Retinopathy Rats And Its Mechanisms

Objective:To investigate the effects of TAP on the experimental diabetes rats nephropathy and retinopathy and its mechanisms.Methods:(1)The Diabetic rats were induced by Streptozotocin(STZ).We observed the influence of the complex prescription TAP and its two composed simple recipe T and P on diabetic rats blood glucose,body weight(BW),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),serum fructosamine(FMN) and blood fat metabolism.(2)The treatment with TAP for 16w,20w and 24w,we collected the urine aliquot(UA)to observe the influence of the drugs on the urine volume(UV),urine protein(UP)and urinary albumin(UAIb)excretion of diabetic rats.When the administration was over,we calculated the kidney index(KI),detected serum creatinine(Cr)and blood urea nitrogen(BUN),nitrogen monoxidum(NO) contents.After making nephridial tissue microtome section,we analyzed the Correspondence Glomerular volume(CV_G)and indices for tubulointerstitial injury(TII) via using computer image analysis software.Meanwhile we used the electron microscope to observe the changes of nephridial tissue ultrastructure.To investigate the protective mechanisms of TAP on diabetic rats kidney,we used the immunohistochemistry to detect the expression of vascular endothelial growth factor (VEGF),transforming growth factor-β1(TGF-β1)and typeⅣcollagen(CO-Ⅳ)in the nephridial tissue.(3)After administering TAP 24weeks,we maked retinal vessel digesting stretched preparation,and used the PAS-H dyeing to analyse capillary vesse endotheliocyte(E),pericyte(P)and E/P.Then we observed the protection of TAP on the retina of diabetic rats via detecting the expression of VEGF by immunohistochemistry.Results:(1)TAP can decrease the content of serum glucose,MDA,TG,TC,LDL-C and increase BW,SOD,GSH-Px and HDL-C of diabetic rats induced by STZ significantly.T and P had also some effects which however were inferior to the TAP group(P＜0.01).(2)TAP can decrease Cr,BUN(P＜0.05,P＜0.01),UP,UAlb(P＜0.001,P＜0.01)of diabetic rats induced by STZ significantly,we can see from the analyzing of the sections that the TAP can decrease CV_G(P＜0.01)and lessen TII (P＜0.001)significantly.TAP can inhibit the expression of VEGF,TGF-β1,CO-Ⅳfrom the results of immunohistochemistry(PΙ0.001).TAP,T and P had protective effects on diabetic rats kidney and TAP was the most powerful(3)TAP can protect pericyte, diminish retinal vessel endothelioeyte exceptional proliferation,degrade E/P,inhibit the expression of VEGF.T and P also had some determinate effect.Conclusion:1.TAP can decrease the serum glucose,regulate the blood lipid disorder and improve the antioxidant ability siginificantly.2.The protective mechanisms of TAP on the kidney of diabetic rats maybe via diminishing the expression of VEGF,TGF-β1,CO-Ⅳof nephridial tissue.3.The protective mechanisms of TAP on the retinopathy of diabetic rats maybe via regulateing the blood lipid disorder,improving the antioxidant ability and diminishing the expression of VEGF in the retina.4.The results of this article illustrate the TAP has a better protective effects than the simple recipe of the T and P.
