Objective To investigate the expression levels of osteopontin (OPN) in the tissues of non-small cell lung cancer and their clinical significance.And the clinical significance of OPN as a tumor marker in peripheral blood of patients with non-small cell lung cancer and the relationship among them.Methods (1)Expression of OPN and MMP9 were detected in all 48 specimens of non-small cell lung cancer , 29 specimens of paracancer tissues and 13 specimens with benign pulmonary disease by tissue microarray and immunoradiometric assay.The interrelations and relationship with the pathological characteristics were analyzed.(2) Plasma levels of OPN and serum levels of CEA were detected in 50 cases with non-small cell lung cancer,15 patients with benign pulmonary disease and 15 healthy controls by ELISA and RIA methods.Results (1)The expression levels of OPN and MMP-9 protein in NSCLC were obisously higher than lung benign disease and adjacent non-cancer tissue(P<0.05),but no significant correlation was detected in lung benign disease and adjacent non-cancer tissues(P>0.05).(2)The expression levels of OPN and MMP-9 in NSCLC were significantly correlated to histological classification,TNM stage and lymph node matastasis(P<0.05),but not correlated with gender,age, size of tumor,cell differentiation and distant metastasis(P>0.05).(3)There was a positive correlation between OPN and MMP-9 expression in NSCLC tissues(r=0.58).(4)The plasma levels of OPN and the serum levels of CEA in group of NSCLC were higher than those in groups with benign pulmonary disease and health(P<0.05),but there were no significant difference between benign pulmonary disease and health group(P>0.05).(5)The plasma levels of OPN were statistically correlated with lymph node metastasis and TNM stage (P<0.05),but not correlated with gender,age,histological type and distant metastasis(P>0.05)in NSCLC group.(6) The serum levels of CEA were correlated with histological type, lymph node metastasis and TNM stage(P<0.05),but not correlated with gender,age and distant metastasis(P>0.05) in NSCLC group. (7)The diagnostic sensitivity(%) of OPN and CEA for NSCLC were 78.0% and 62.0%;the diagnostic specificity(%) of OPN and CEA for NSCLC were 88.7% and 93.3%, no statistically significance among them (P>0.05). The sensitivity of combined with two markers detection was 90.00%.Conclusion (1) The overexpression of OPN and MMP-9 were closely correlated with invasion and matastasis in NSCLC. Combined detection of OPN and MMP-9 may be helpful to determine metastasis and staging of NSCLC. (2) OPN and MMP-9 were over-expressed in NSCLC tissues and accordant among them.They might play synergetic role in carcinogenesis and progression of NSCLC.(3) Combined detection of plasma OPN and serum CEA can signicantly improve the diagnostic sensitivity and specificity in NSCLC.
