ObjectiveTo observe the expressions of KL-6 ,IL-4 and IFN-γin serum and bronchoalveolar lavage fluid in asthma mouse model, and study the effects of bacillus calmette-guerin(BCG) treatment on them. To understand the relationship between KL-6 and asthmatic airway inflammation and the mechanism.Methods50 male Kunming mice were randomly divided into five groups, with 10 mice in each group.①Asthmatic groupⅠ: each mouse was sensitized by intraperitoneal injection 10ug ovalbumin(OVA) and 1mg aluminum hydroxide, and challenged by inhaling 1%OVA aerosol for 3 days.②Asthmatic groupⅡ: each mouse was sensitized and challenged in the same methods as asthmatic groupⅠ, but inhaling 1%OVA aerosol for 7 days.③Asthmatic groupⅢ: each mouse was sensitized and challenged in the same methods as asthmatic groupⅠ, but aerosol 1%OVA for 11 days.④BCG treatment group: each mouse was sensitized and challenged in the same methods as asthmatic groupⅡand was intradermal injection with BCG 0.025mg on 7,3,1 day before sensitization.⑤Control group: mice was sensitized and challenged with NS. Twenty-four hours after the final challenge, serum and bronchoalveolar lavage fluid(BALF) were collected. The total number of cells and eosinophils in BALF was counted. The levels of KL-6,IL-4 and IFN-γwere determined using Enzyme-Linked Immunosorbent Assay(ELISA). Sections from lung tissue were stained with HE.Results1. There are many inflammatory cells to infiltrate lung tissue in each asthmatic group, the more the mice inhale the more the inflammatory cells infiltrate.2. The number of total cells and eosinophils in BALF of each asthmatic group was more than that of control group. With the time of OVA inhaled prolonging, the number of total cells and eosinophils in BALF rised.3. The level of KL-6 in serum and BALF of each asthmatic group was more than that of control group. With the time of OVA inhaled prolonging, the levels of KL-6 in serum and BALF rised.4. Compared with control group, the expressions of KL-6 and IL-4 in BALF increased significantly in asthmatic group and BCG treatment group, but the expressions of KL-6 and IL-4 in BCG treatment group were lower than that in asthmatic group. The expression of IFN-γin BALF decreased significantly in asthmatic group and BCG treatment group than that in control group, and increased in BCG treatment group compared with asthmatic group.5. A positive correlation between KL-6 level and IL-4/IFN-γoccured in asthmatic group and BCG treatment group.Conclusion1. The expression of KL-6 can be found in serum and BALF in asthmatic mice model, and KL-6 can reflect a positive correlation with airway inflammation .2. The expression of KL-6 in BALF in asthmatic mice model can be reduced by BCG. There is a positive correlation between KL-6 and IL-4/IFN-γ. The expression of KL-6 related the type 2 T helper domination.3. KL-6, just like cell adhesion molecule, plays an important role in asthma.
