Objective: This study was designed to primarily investigate the inhibitory effects and phase growth arrest of sodium phenylbutyrate(PB) on gastric cancer cell line SGC-7901 in order to acquire more information about PB functions on the treatment of gastric cancer.Methods: MTT(Methy thiazolyl tetrazolium) assay was used to evaluate the inhitory effects of PB in different concentration and time. Alteration of the morphology of PB induced on SGC-7901 cell line in vitro was observed by invert microscope. FCM(the fluorescence flow cytomtry) DNA assay were applied to detect the alteration of cell-cycle phase. Also MTT was used to examine the growth inhibition of SGC-7901 cells treated by the combination of PB with 5-FU.Results: PB could inhibit the growth of SGC-7901 and the inhibitory effects showed time-and dose-dependent pattern, the difference shows significant ( P<0.05 ) .Morphological changes were observed by invert microscope. FCM assay indicated that most of the cells were arrested in G 0/G1 phase. We found the drug combination of PB and 5- FU can increase the inhibit rate of SGC-7901 cell, compared with 5-FU only,the difference shows significant(P<0.05).Conlusion: PB can inhibit the growth of the gastric cancer cell line SGC-7901 cells in vitro. The inhibitory effect of sodium phenylbutyrate on SGC-7901 may through the increasing of G0/G1 fraction with a concomitant drop in the S-phase.The drug combination of PB and 5-FU can increase the inhibit rate of SGC-7901 cell, compared with 5-FU only.
