Cervical cancer and gastric cancer were charactered with high incidence rate, as well as difficult therapy. During the chemotherapy process, anticancer agents were harmful to normal cells. This drawback restricts the broad application. Therefore, it is urgent to develop a new type of compounds with high-efficacy and low-toxicity to increase susceptivity of chemotherapy agents or to reduce the toxicity of the chemotherapy agents. In this paper, we investigated that the growth inhibition activity of Allium Chinese extraction against Hela cells and BGC-823 cells. Futhermore, we studied the boosting effect of Allium Chinese extraction on 5-FU for killing Hela cells and BGC-823 transforming cells. And the difference between the proteins. Expressing in cells dealt with Allium Chinese extraction and that in normal cells was compared by IDE-MS/MS, to analyze the mechanism of Allium Chinese extraction in increasing efficacy and decreasing toxicity of 5-FU.The inhibition rate of cell proliferation was measured by MTT method. And the apoptosis of BGC-823 cells and Hela cells was observed by Fluorescent microscope. At the same time DNA fragments of apoptosis cells were analyzed by DNA ladder method. The results showed that the bioactive components of Allium Chinese markedly inhibited the proliferation of BGC-823 cells and Hela cells, in a concentration-dependent manner. Secondly, the growth inhibition activity of bioactive components of Allium Chinese combined with 5-FU is higher than that of single-drug group. We also observed the AO / EB nuclear fluorescence staining the cells after apoptosis Under the microscope DNA fragments were observed by Agarose gel electrophoresis which demonstrated that the bioactive components of Allium Chinese could markedly induce apoptosis of cancer cells.BALB/c mice were injected with the bioactive components of Allium chinese at high,middle and low doses , or co-injected the bioactive components of Allium Chinese combined with 5-FU once a day, lasting for seven days. The effectiveness of the bioactive components of Allium Chinese were evaluated by detecting the immune organs of mice, the phagocytosis of peritoneal macrophage and the activity of lymphocytes. The results showed that 100μL or 150μL bioactive components of Allium chinese could markedly improve the phagocytosis activity of peritoneal macrophage and induce lymphocytes proliferation. We also found that the bioactive components of Allium Chinese at high and middle doses could strongly inhibit the effect of 5-Fu which was referred to doing harm to peritoneal macrophage,lymphocytes and leading to weight loss of mice.From the total proteins of tumour cell dealt with the bioactive components of Chinese Allium , we can' t detect the Prohibitin-2 and Rho-GTPase-activatingprotein4 by SDS-PAGE and MS analysis .This result suggested that the role of bioactive components of Chinese Allium was closely related to p53-induced apoptosis and the regulation of G1/S . Its Molecular mechanism needs further study.
