The aim of this paper is to investigate the influence of ethanol extract of the allium (EA)on immune function in mouse.60 Kunming mice were randomly divided into the cyclophosphamide group (C), cyclophosphamide + alcoholic extract group(C + A), ethanol extract of the allium group (A) and control group (S). The mice of C group and C + A group were subcutaneous injected cyclophosphamide to induce immune suppression; the mice of C + A group and A group were fed with EA for 35 days; the mice of C group and S group were fed with physiological saline. The immune organ index, pathology change, the cellular and humoral immune function and the non-specific immune function were measured by the DTH , flow cytometry,HC50 assay and conventional methods. The results as follow:1. Ethanol extract of the allium promoted weight gain in mice.The weight gain of A group was the highest in the four groups (P<0.01). The weight gain of S group was significantly higher than that of the C group and C+A group (P<0.01) on 1 week post treatment.There was no significant difference between C group and C+A group on 1 week(P>0.05). The weight gain of A group and S group was significantly higher than that of the C group and C+A group in 2 week to 5 week(P<0.05).2. Ethanol extract of the allium increased immune organ index .The thymus index of C group was the lowest in the groups (P<0.01), The thymus index of C+A and A group was significantly higher than that of S group (P<0.01).There was no significant difference between A group and S group in spleen index (P>0.05),but C+A group and A group was significantly higher than that of the C group(P<0.05).3. Ethanol extract of the allium enhanced the specific immunity function.T cell activity,peripheral blood T cell subgroup and B cell activity were detected to assay the effect of EA on the specific immunity function.The weight difference of the left and right ear of C+A group,A group and S group was significantly higher than that of the C group(P<0.05). The weight difference of the left and right ear of the C+A group was higher than that of the S group (P<0.01). There was no significant difference between A group and S group (P>0.05). The CD4+/CD8+ values of C+A group,A group and S group were significantly higher than that of the C group(P<0.01),the C+A group was higher than A group and S group(P<0.01). There was no significant deviation between A group and S group in CD4+/CD8+values (P>0.05).The serum antibody of C group was the lowest in the groups (P<0.01).The serum antibody of C+A group was significantly lower than that of S group (P<0.01).There was no significant difference between A group and S group in serum antibody (P>0.05).4. Ethanol extract of the allium enhanced the no-special immunity function.The activity of peritoneal macrophage phagocytic and NK cell were detected to assay the influence of EA on the non-specific immunity function.The peritoneal macrophage phagocytic activity of C+A group and A group was significantly higher than that of C group and S group (P<0.01).The NK cell activity of C group was significantly lower than that of the other groups (P<0.01).The NK cell activity of the A group was higher than that of C+A group and S group (P<0.05).5. Ethanol extract of the allium improved the Pathology lesion.Liver cell cords of the C group mice were inordinate, focal degeneration and necrosis appeared in liver cells. The germinal center of the lymph node was obviously. While the spleens of C group were atrophic, containing less lymph-cell. The thymus cortex of the C group was significantly thin .Structure of liver cell cord of C+A group becamed clear, the germinal center of C+A group was not obvioused. There were many macrophage and polynucleation macrophage in the spleen of C+A group.There were a lot of lymph cell in cortex of thymus.Liver cell cords of the A group mice were clear, lymphatic nodules and the germinal center is obviously. There were more macrophage in the spleen of A group. The cortex of thymus is plenty.Liver cell cords of the S group mice were clear, the germinal center of S group was obvioused. There was few macrophage in the spleen of S group. The cortex of thymus was plenty.6. ConclusionThe results showed that EA enhanced the weight gain and immune organ index; increased the weight difference of the left and right ear, CD4+/CD8+values, serum antibody and phagocytic activity in immune suppression mice. As the same time, EA promoted phagocytic activity and NK cell activity in the normal mice. So the conclusion can be obtained that EA can significantly improve the specific immunity and non-specific immune function in immune suppression and normal mice.
