Objective: To evaluate the in vitro biological effects of nerve growth factor and recombinant human bone morphogenetic protein by acting individually or jointly on the proliferation and alkaline phosphatase activity of human periodontal ligament cells . The purpose of this study is to applies for the growth factor in the clinical periodontosis's treatment provides the rationale.Methods: (1) Human periodontal ligament cells were cultured by tissue explant . (2) To find out effects of nerve growth factor and recombinant human bone morphogenetic protein by acting individually or jointly on the proliferation and alkaline phosphatase activity of human periodontal ligament cells, its proliferation were mesured by MTT colorimetric assay, ALPase activity was measured by enzyme kinetic methods. (3) Observe dynamically the effects of nerve growth factor and recombinant human bone morphogenetic protein by acting individually or jointly on the proliferationof human periodontal ligament cells.Results : (1)Either NGF or rhBMP can promote the proliferation of human PDLCs(P<0.05), the optimal density of NGF is 10U/ml; the optimal density of rhBMP is 200ng/ml.Significantly greater proliferation was found in NGF(10)+rhBMP (200) (sum of the optimal density) compared with individual action(P<0.01). (2) The results shown NGF and rhBMP can promote alkaline phosphatase activity of human periodontal ligament cells by acting individually or jointly(P<0.05). Significantly greater promote was found in NGF(10)+rhBMP (200) (sum of the optimal density) compared with individual action (P<0.01). (3) NGF and rhBMP by acting individually or jointly on the proliferation of human PDLCs , observe dynamically from 1 to 9 days and their role is not promoting proliferation trends show continuous increase.Conclusions: Either NGF or rhBMP can promote the proliferation and alkaline phosphatase activity of human PDLCs, and combined two actions had synergistic function; NGF(10)+rhBMP (200) was optimal density. Observe dynamically the effects of NGF and rhBMP by acting individually or jointly on the proliferation of human PDLCs from 1 to 9 days, their role is not promoting proliferation trends show continuous increase. NGF and rhBMP can be used as bioactive mediator on periodontal regeneration.
