Objective:Laryngeal carcinoma is one of malignant tumors common in head and neck,its mainly pathologic type is laryngeal squamous cell carcinoma,account of 95%-99%.Its etiopathogenisis is complicated and incidence is growing up.Human epidermal growth factor receptor(EGFR) is one member of protein tyrosine kinase receptor family,its abnormality may promote the growth,migration,vasiformation of tumors and supress the apotosis of tumor cells.The purpose of this experiment is to detect the amplification and overexpression of EGFR gene in laryngeal squamous cell carcinoma and and investigate their clinical significance.To illustrate the molecule mechanism of laryngeal carcinoma genesis and development,provide evidence for clinical diagnosis and progonosis.Methods:EGFR gene amplification in 40 cases carcinoma tissues and paired normal tissues was detected by differential PCR approach.EGFR mRNA were evaluated by reverse transcription polymerase chain reaction(RT-PCR).Statistical analysis was performed using SPSS13.0.Analyzed the difference of EGFR amplification and overexpression,the correlation between amplification and overexpression in 40 cases carcinoma tissues and normal.Analyze the association between them and clinical parameters of 40 patients,such as age,sex,the size of tomer,differentiation of tumor cells.Results:1.The picture of gel was taken by UVP gel imaging system.Measuring the integrated optical density of these electrophoresis strips with the same software.In the 40 LSCC cases,EGFR amplification was deteted in 13 patients(32.5%),but negative in paired normal tissue.Analyzing these data with SPSS13.0,EGFR amplification of the tumor group is obviously higher than that group of normal control(x~2=15.537,P<0.001).2.EGFR mRNA is overexpression in laryngeal squamous cell carcinoma.The mean integrated optical density of EGFR is 872.356±62.340 in laryngeal carcinoma tissue,346.425±57.380 in the paired normal tissue(t=5.959,P<0.05).Using EGFR upregulated by 30%as our standard,the overexpression was detected in 29 cases of the LSCC patients(72.5%)(x~2=45.490,P<0.001).3.Through correlation analysis,it was found that there was a high consistency between PIN1 gene amplification and mRNA overexpression in laryngeal carcinoma tissue.Statistical analysis showed that there was a significant correlation of EGFR amplification and mRNA overexpression with advanced clinical pathological stage,and also with poor differentiated degree of cancer cells(P<0.05)Conclusion:This research results suggested that EGFR gene amplification contributes to its mRNA overexpression.EGFR gene may play an important role in laryngeal carcinoma oncogenesis and malignant progression.Therefore,EGFR may predict outcome in laryngeal Carcinoma patients and provide useful information for clinical treatment. |
