Cancer is a serious hazard to human health major refractory disease, and drug therapy has an important position in three treatment of malignant tumors. There are many antitumor drugs developed or developing at home and abroad, but few are efficient or low toxic side-effect. Therefore this is a research point to continue looking for the highly efficient and low toxicity antitumor drugs. China has rich resources of medicinal plants, and the antitumor drug research and development projects are keeping endless in natural plant since the 1970s. Many papers also reported that most of antitumor synthetic chemical drugs are based on natural antitumor constituents of the compounds, including computer simulation and design are also bionic natural antitumor constituents-based. Generally, plant sources of antitumor drugs have low side-effect, so it will be broad prospect to develop independent intellectual property rights of antitumor drug from natural medical plant sources, modern medical analysis pointed out that saffron (Crocus sativus L.), which was a traditional Chinese medicine, contained crocin, crocetin, safranal, and other important secondary metabolites, which all had significant antitumor or tumor inhibition abilities. However, they are all contained in stigmas, and have very low yields. The saffron corm takes the majority part of the harvest weight, and this prompt us to find ways to separate similar antitumor materials in corms.Escribano(1999) isolated a new type of proteoglycan from saffron corm the first time, and research shows its strong cytotoxic inhibition for HeLa cells. In this project, we took a series of experiments restructuring and improving basing on their papers. In DEAE chromatography extraction, we extended the length of the chromatography column (16 mm×200mm), evaluated and optimized the chromatographic separation from the sample, flowing rate, detection method, and separation efficiency to increase the production significantly. Later separation used HPLC, analyzed it from eluted method, separation efficiency, and degree of separation material. The inhibition rates of each fraction were established by HeLa cells and A549 cells. The separation rate in chromatography was 0.231%, in HPLC was 19.3%, and the final separation rate was 0.045%.The results of the subject show that the active saffron corm material may be a valuable antitumor drug, and initially establish an extraction method of the active saffron corm antitumor material. This will make feasibility models and support data for the following large-scale experiment of industrial production.
