Objective:To establish simple and effective method for rapid identification of Crocus sativus L. and determination of the content of crocusⅠand crocusⅡin Crocus sativus L. Providing a system scientific method for scientific evaluation and effective control of the quality of Crocus sativus LMethods:Maroscopic,microscopic and TLC traditional identification methods, combining GC method were performed to identify Crocus sativus L. and its counterfeits; Determination of the content of crocusⅠand crocusⅡin Crocus sativus L:Chromatographic conditions included column Agilent ZORBAX StableBond SB-Aq (4.6 mm X 250 mm,5μm) with 0.01%formic acid (A)-acetonitrile (B) as the mobile phase gradient elution. 0 min:75%A; 8 min:70%A; 10 min:60%A; 25 min:30%A. The flow rate was 1.0 ml/min, the injection volume was 5μl, the detection wavelength at 440 nm, the column temperature was 30℃. Result:1. On properties:There were certain differences between Saffron and its adulterants. From outside, the fake Saffron looks better than Safflower, But the hand would be stained in red or felt oil when rubbing them. Judging from the smell, the Saffron smell peculiar, have a little pungent odor and flavor bitterness. But the other were odourless or smelled like vegetable oil.2. From microscopic observations of powder, the difference between Safflower and its adulterants was great. Pollen grains of saffron was unique and the outside of the epidermis in projection protrude papillary or villous. All of these features could not be found in the fake Saffron.3. The results of GC identification showed that Chromatographic graphics of the Saffron and its fakes were markedly different; There were also different between quality goods and adulterate Saffron, retention time of characteristic peaks were same, but the peak heights is distinct.4. The results of the determination of the content of crocusⅠand crocusⅡin Crocus sativus L. showed that the retention time of crocusⅠand crocusⅡwas about 5.71min and 8.83min respectively. The regress equation for crocusⅠwas y= 24.928x-0.2543,r=1, and the linear range was 3.125-200μg/ml. That for crocusⅡwas y= 36.106x+8.3698,r=1, and the linear range was 3.125-200μg/ml.The average recovery of crocusⅠand crocusⅡwas 100.5% and 100.2%, RSD was 1.43% and 1.4% respectively.Conclusions:1. The traditional methods of Maroscopic,microscopic and TLC identification were simple and reliable, and can be used for initial identifying Crocus and its counterfeits.2. Using gas chromatography can make a further identification for Crocus sativus L.. This is a simple and accurate reproductive method which can be used for the identification of Crocus sativus L.3. The method is accurate and sensitive. It can be used for the content determination of crocus I and crocus II in Crocus sativus L. from various places of china.
