New modern medicine theory thinks that many diseases are caused by excessive free radicals of human body. Diseases that related to free radical now includes inflammation, cardiovascular disease, senescence, tumour, ischemic disease, poisoning and radiation sickness and so on. In recent years, as a new direction of chemistry and medical science researches on free radical and its scavengers and dynamic mechanism gradually attracted more and more attention and got many research results .With the in-depth research of antioxidant, people found that Synthetic antioxidant have a lot of disadvantages, such as poisonous side effect , bad heat stability , low antioxidant efficiency ,narrow apply range , low bacteriostatic activity , natural antioxidant become the main current of antioxidant for its incomparable advantage such as natural, high efficiency and low poisonous side effect. Researches on the antioxidation effects of aromatic plants extracts have becoming the trends of international and domestic research, However, most studies were focused on essential oils. The studies on the nonvolatile parts are limited, especially on their abilities for scavenging free radical , antioxidation and protecting oxidative damaged lung cells, most of which are about well known aromatic plants, such as lavender and rosemary.In the present study the nonvolatile parts of dry leaves from Pelargonium graveolens were taken by using three extraction methods. And obtained three different extracts, which were used as the experiment materials,Six chemiluminescence systems and two colorimetric systems were used to systemically evaluate their capacity of scavenging all kinds of free radical ( ROS , RNS , RNC), inhibiting lipid pexoxidation and preventing DNA oxidative damage caused by OH. The effect and mechanism of nonessential oil extracts of Pelargonium graveolens protecting cultured HELF cells from oxidative damage were also studied to evaluate their anti-athrosclerosis activities.The experiments revealed that the components of nonessential oils in Pelargonium graveolens leaves could not only directly scavenge O2-, but also could efficiently scavenge·OH , H202, ONOCT, C0-·3 and DPPH·, inhibit lipid peroxidation and obviously inhibit and delay DNA damage caused by OH, acting as mixing-type antioxidants.The results of cell experiments indicated that water and ethanol extractions not only can promote prominently proliferation of HELF on low concentration (1-10μg/mL) while inhibit proliferation of HELF on middle or high concentration (>10μg/mL) and influence cell cycling phase , but slao can dose-dependently increase the activities of SOD , CAT and the level of GSH , inhibit the production of 0N00-. And MDA in HELF or in the medium of the HELF while cultured HELF were damaged by H2O2. Above studies revealed that the components of nonessential oils in Pelargonium graveolens leaves can increase the oxidase activities and scavenge ROS and RNS directly, possiblely can prevent lung tissue from oxidative damage, reduce production of LPO and alleviate or prevent lung diseases.The results also showed that the components of nonessential oils in Pelargonium graveolens leaves all contain certain flavonoids and phenols. The antioxidative abilities were correlated with their content of flavonoids, which suggested that flavonoids in Pelargonium graveolens leaves are the effective components.Above results indicate that the components of nonessential oils in Pelargonium graveolens leaves are very effective and multifunctional antioxidants and free radical scavengers with a good application prospect. The present study developed the application foreground of Pelargonium graveolens and improve its utilization ratio and especially establish theoretic base for the function study of Pelargonium graveolens on medical treatment and nutrition research field.
