Radix Astragali, Huangqi in Chinese, the dried root of Astragalus membranaceus (Fisch) Bge. Var. mongholicus (Bge.) Hsiao or A. membranaceus (Fisch) Bge., is one of the most widely used Chinese herbs prescribed in many Chinese formulas to reinforce "Qi" (vital energy). It was used for the treatment of nephritis, diabetes, hypertension, cirrhosis, cancer etc., as an important tonic, an antiperspirant, an atonic, a diuretic and an adjunctive medicine by Chinese doctors. As Traditional Chinese Medicine (TCM) of tonic function, Radix Astragali has been used thousands of years. The quality of TCM can be evaluated by the chromatographic fingerprints, which represent the compound groups of TCM and substantial foundation of pharmacodynamics. The chromatographic fingerprints can cater to the theory of Chinese traditional medicines and characteristics of chemical constituents in TCM, thereby also reflect the quality of TCM in an all-around way and be accepted by international. The article established the chromatographic fingerprints of Radix astragali and preparations with HPLC/DAD.As reported previously, the chemical comprises of Radix astragali mainly includes polysaccharides, flavonoids and saponins, among which saponins are commonly considered as the major bioactive constituents. The fingerprints of Radix astragali, total Astragaloside and its injection were investigated in this paper, so the saponins of Radix astragali were inspected during the research course of fingerprint.Firstly, the pretreatment conditions of Radix astragali were inspected. By comparing different solvent, extraction method and extraction time, the most optimized extraction condition were ascertained with refluxing extraction with ethanol 30 mins. Then, the chromatographic columns, detected wavelength and reference substance were investigated. The UV absorption was large with stable baseline at 210nm. The Intersil ODS -3 column was chosen as fingerprint analysis column with the wavelength at 210nm. Astragaloside IV was not suitable for reference substance because the peak of it was very small or not obvious. Clinodiside A was ascertained to be reference substance and mixed by autosampler of the HPLC instrument. The fingerprint analysis method of Radix Astragali from GAP producing area was developed, and the methodology including precision, stability and reproducibility were inspected. There were eight peaks in the chromatogram including clinodiside A. The fingerprints of the raw material of total Astragaloside and its injection were also developed based on the research of Radix astragali. The developed methods could be used to control quality of the medical material and its perparations.By many kinds of separation methods, 12 compounds were isolated from the raw material of total Astragaloside. Their structures were elucidated based on spectra data (1H-NMR, 13C-NMR, DEPT, 1H-1HCOSY, NOESY, HMQC, HMBC, EI-MS, ESI-MS, IR).
