Objection:To investigate the suppression of SGC7901 cell line by transfecting combined p53 and AS genes ,explore the potential of combined gene therapy in the treatment of human gastric cancer.Method:The pVITRO2-p53-AS was transfected into human gastric cancer cell line SGC7901 with liposome and paralleled with the vector control and the p53,AS contro1.Gene transcription was examined by RT—PCR.Cell growth and apoptosis were observed by cell clones trial, MTT growth curve , flow cytometry detecting cell cycle and Annexin V .Statas were analyzed by SPSS10.0.Results:1,RT-PCR:The result confirmed the stable expressing of p53 and AS in SGC7901.2,Cell clones trial showed:The number of Cell clones in transfected group was less than that in untransfected group( P< 0. 05 ). The number of Cell clones in combined-gene transfected group was significantly less than sigle-gene transfected group ( P< 0. 05 ).3,MTT growth curve showed:Transfected group showed a significantly decrease in cell growth compared with the vector control group and prolonged in the coupling time ( P< 0. 05 ). The suppressive effect on cell in combined-gene transfected group was stronger than in sigle-gene transfected group ( P< 0. 05 ). 4,Flow cytometry detecting cell cycle:Cells proliferation were hampered in G1/S in transfected groups. The rate of cells in G1 /S of combined-gene transfected group was higher than sigle-gene transfected group( P< 0. 05 ) ,which revealed P53 an AS gene were synergistic to each other on the growth of SGC7901.5,Flow cytometry detecting cell apoptosis and Annexin V:The peak of apoptosis appeared in transfected groups differently. The rate of apoptosis in transfected groups was higher than the vector control group,and combined-gene transfected group was higher than sigle-gene transfected group( P< 0. 05 ) . The Annexin V of transfected group was higher than the vector control group( P< 0. 05 ),and combined-gene transfected group was higher than sigle-gene transfected group( P< 0. 05 ).Conclusion:P53 an AS genes suppressed the growth of SGC7901 by synergistic effect on the apoptosis of cell. It revealed combined gene therapy wa s more powerful than single gene in the therapy of malignant tumor.
