Objective1.To study the expression and significance of membrance-bound Platelet-derived growth factor receptors( mPDGFR-É‘and mPDGFR-?) in preeclampsia, as well as the possible pathological and physiological roles of mPDGFR in preeclampsia.2.To study the relationship between the soluble Platelet-derived growth factor receptors'concentration in maternal peripheral vein blood and the preeclampsia. And investigate the relationship with Severity of the cases.3.Analyze the relationship between mPDGFR expression and clinical parameter, such as MAP, neonate birth weight , 24h Urine protein and placental weight.MethodsThe expression of mPDGFRs in the placenta of the preeclampsia group and the control group was detected by Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry test(IHT). Moreover, we assay sPDGFRs on serum of maternal peripheral blood using enzyme-linked immunosorbent assay(ELISA).Results1.The expression of both mPDGFRs mRNA and protein in the preeclampsia group was higher than that expression in the control group (P<0.01).2.the Serum level of sPDGFRÉ‘and sPDGFR-? was lower in the preeclampsia group than that in the control group(P<0.01).3.According IHT, we found that mPDGFR-É‘was mainly expressed in decidual cells and syneytiotrophoblasts, less expressed in vascular endothelial cells , chorionic interstitial cells and so on;While mPDGFR-? was mainly expressed in chorionic capillary endothelium, less expressed in chorionic interstitial cells, chorionic interstitial cells and so on.4.The expression of mPDGFRs in placenta has a significant positive correlation with MAP and urine protein, and a significant negative correlation with neonate BW and placental weight.Conclusion1.The high expression of mPDGFR-É‘mRNA and protein in the preeclamsia suggests that mPDGFR-É‘may participate in the pathophysiological process in the preeclampsia;They may influence the differentiation and infiltration of nutrient cells, and then induce superficial nidation of the placenta in the preeclampsia.2.The high expression of mPDGFR-? mRNA and protein in preeclamsia suggests that mPDGFR-É‘may participate in the pathophysiological process in the preeclampsia;They may participate in the atherosclerotic changes of placenta vessels in the preeclampsia.3.The low serum level of sPDGFRÉ‘and sPDGFR-? in the preeclamsia suggests that sPDGFR may preserve women from preeclampsia.
