Objective:To make"The Compound Danshen Sustained-release tablet"as prototype,studied the extraction and treatment technology of major material Danshen root in the tablet.and studied the release degree of The Compound Danshen Sustained-release tablet which is produced by our laboratory and the material is made by ourselves.the study supplied a new method for research of the Compound Danshen Sustained-release Preparation.Method:Tanshinone-ⅡA and salvianolic acid B are both inthermostable,when heating long time,they can breakdown quickly.so it's difficult in actually industrial production .the study makes Tanshinone-ⅡA and salvianolic acid B as evaluate index,take HPLC method to determine the contents. According to the monofactorial test and orthogonal test To Investigate the influence of such items as extration temperature,extraction solvent,the volume of solvent and extraction times .optimized the best method of Tanshinone-ⅡA's extraction and salvianolic acid B's extraction technology.the paper introduced a technology which can extract Tanshinone-ⅡA and salvianolic acid B at the same time,and supplied the combination extraction technology.so the paper researched the treatment method of the extraction liquid,and decided a technology which can purified the extraction liquid,and make the value component have the lest waste. The paper through system methodology research to established a method which fit for determine the release of Compound Danshen Sustained-release Preparation in vitro,the Compound Danshen Sustained-release Preparation is made by our liberatary,and the extraction of danshen and sanqi are also supplied by ourselves.Determined the release of indexes components in water by HPLC,and make this to explain the release mode in vitro of the index of Danshen in Compound Danshen Sustained-release Preparation.Compared the Compound Danshen Sustained-release Preparation which made by our liberatary and the Sustained-release Preparation which suppliede by meichen company,evaluated the release degree of two Sustained-release Preparation.Results:The new idea of the study is introduced a technology which can extract Tanshinone-ⅡA and salvianolic acid B at the same time,and supplied the combination extraction technology:14 times extrating solvent1,make the particular solvent 1,extraction 150minutes(the first time is 90minutes,the second time is 60minutes) under 80℃.the outcome indicated,under this technology,the content of salvianolic acid B in extraction liquor is 75.23mg/g,the diversion rate is 95%; the content of Tanshinone-ⅡA in extraction liquor is 2.763mg/g,the diversion rate is 91.49%.At the same time,it studied the treatment of extraction liquor,and introduced the isolation and purification technology of Tanshinone-ⅡA and salvianolic acid B .Researched the influence of concentration technology, concentration ratio, precipitation technology and the vacuum drying temperature,in the end,we supplied the best technology:concentrated the extraction liquid to ratio of 1:1 under reduced pressure, vacuum filtrated, reserved the filter residue, the filter liquor was precipitated with ethanol to the ethanol content is 80%,take the precipitated liquor and concentrated dense, vacuum dried under 50℃.the result showed,the content of Tanshinone-ⅡA in dried solid is 2.54mg/g,the aversion rate is 84.27%;the content of salvianolic acid B in dried solid is 69.49mg/g,the aversion rate is 87.75%,the yield of solid is 19.23%。the study result showed,the method is better than the literature's report,in the end,the aversion rate of active components is very high,the yield of solid is very low,and the method is simply,conveniently,and can be used for industry production. The paper through system methodology research to established a method which fit for determine the release of Compound Danshen Sustained-release Preparation in vitro. It determined the release of indexes components in water by HPLC,and make this to explain the release mode in vitro of the index of Danshen in Compound Danshen Sustained-release Preparation.The result shows , main indexes salvianolic acid B and tanshinoneIIA release balance behavior in 2-10 hours. The release curve is near to the Higuchi equation's release mode. Compared the Compound Danshen Sustained-release Preparation which made by our liberatary and the Sustained-release Preparation which suppliede by MeiChen company,evaluated the release degree of two Sustained-release Preparation.Conclusion:The study introduced a technology which can extract Tanshinone-ⅡA and salvianolic acid B at one time,the outcome indicated,under this technology,the content of salvianolic acid B and Tanshinone-ⅡA in extraction liquor is highly;The paper introduced the isolation and purification technology of Tanshinone-ⅡA and salvianolic acid B .researched the influence of concentration technology,concentration ratio,precipitation technology and the vacuum drying temperature,in the end,we supplied the best technology,and the result showed,the content and the aversion rate of Tanshinone-ⅡA and salvianolic acid B in dried solid is highly,the yield of solid is lowly;the decided of extracted and treated technology give us a new method for danshen's extracting and treating.The paper through system methodology research to established a method which fit for determine the in vitro release of Compound Danshen Sustained-release Preparation.The paper compared the Compound Danshen Sustained-release Preparation which the material is supplied by our liberatary and the Sustained-release Preparation which the material is supplied by MeiChen company,evaluated the release degree of two Sustained-release Preparation. Established a method which can be used to evaluate the release of Compound Danshen Sustained-release Preparation,the research supplied a new idea for study of the Sustained-release Preparation.
