Objective:To investigate the relationship of the expression of LaminB1 and NMP238 in pre-radiotherapy tissue with the radiation sensitivity and prognosis in human nasopharyngeal carcinoma tissue.Methods:1.52 samples from patients with pathologically diagnosed as nasopharyngeal low-differentiation squamous cell carcinoma were collected.These patients had done some examinations before treatment and demonstrated that there was no distant metastasis.All cases had completed the radical radiotherapy.52 patients were classified into two groups according to the response to radiotherapy as complete response (CR)and partial response groups(PR).2.Immunohistochemical methods were performed to detect the expression levels of LaminB1 and NMP238 in pre-radiotherapy tissue.3.The positive expression rates of LaminB1 and NMP238 in nasopharyngeal carcinoma tissue were observed.The potential relations of expression of LaminB1 and NMP238 with clinic stage,radiation sensitivity and prognosis were also analyzed. Results:1.The positive expression rates of LaminB1 and NMP238 in nasopharyngeal low-differentiation squamous cell carcinoma were 79%,67%respectively.2.The expression intensities of LaminB1 and NMP238 in nasopharyngeal carcinoma significantly related with their clinical stages(P<0.05).3.The expression of LaminB1 in nasopharyngeal carcinoma tissue was positively correlated with that of NMP238 by Pearson correlation analysis(C=0.446,P<0.05).4.In 52 cases,only 36 cases(69%)were complete response,16 cases(31%)were partial response,while no change and progress disease were 0 and 0 respectively.5.According to the expression intensity of LaminB1(-~+++), the complete response rates were 81.8%(9/11),79.3%(23/29),37.5%(3/8) and 25.0%(1/4)respectively,and there was a significant correlation in them(P<0.05).According to the expression intensity of NMP238(-~+ ++),the complete response rates were 82.3%(14/17),77.3%(17/22),33.3%(3/9)and 50%(2/4)respectively,and there was a significant correlation in them(P<0.05).6.In 52 cases,the 5-year survival rates in stagesⅠ~Ⅱ,Ⅲ,ⅣA were 84%,53%,20%respectively. 7.According to the expression intensity of LaminB1(-~+++), the 5-year survival rates were72.7%(8/11),72.4%(21/29),25.0%(2/8)and 25.0%(1/4)respectively,and there was a significant correlation in them(P<0.05).According to the expression intensity of NMP238(-~+ ++),the 5-year survival rates were 82.3%(14/17),68.2%(15/22),22.2%(2/9)and 25%(1/4)respectively,and there was a significant correlation in them(P<0.05).Conclusions:1.There was a correlation between the expression intensities of LaminB 1 and NMP238 in nasopharyngeal carcinoma and their clinical stages.2.There was a co-relationship between the expression of LaminB1 and NMP238 in nasopharyngeal carcinoma.3.The expression intensities of LaminB1 and NMP238 significantly related with radiation sensitivity and prognosis.It suggests that the expression levels of LaminB1 and NMP238 can be as useful predictors for the radiosensitivity and prognosis in nasopharyngeal carcinoma.
