Objective This present study mainly aimed to investigate the proliferation and apotosis effects of celecoxib in combination with x-ray on human cholangiocarcinoma cell line QBC939and the possible mechanism involved.Methods First, Cell Counting Kit-8(CCK-8) assay to investigate the inhibitive rates after the treatment of celecoxib and x-ray. We determined the IC50by CCK-8assay, which was adopted in the following experiments. Then the cells were divided into five groups:control group, x-ray group, celecoxib group, x-ray after the celecoxib group and x-ray before celecoxib group. Finally, apoptosis of each group were determined by flow cytometry, the protein levels of apoptotic-associated gene survivin was determined by western blotting.Results The apoptosis rates after the celecoxib in combination with x-ray were enhanced apparently to22.133,15.733%from8.267,11.233%(P<0.05). Furthermore, Western blotting showed that survivin protein expression was down-regulated obviously by celecoxib in combination with x-ray (P<0.05). But the efficacy of the x-ray after celecoxib group is better than the x-ray before celecoxib group(P<0.05).Conclusion celecoxib could have the effect of radiosensitization on the QBC939cells. One of the mechanisms is that celecoxib may reduce the expression of survivin or accelerate the degradation of survivin protein.
