Establishment And Study Of Human Oral Verrucous Carcinoma And The Expression Of Survivin And Bcl-2 In Nude Mice Models

Objective:The aim of this investigation was to investigate the establishment experiments in nude mice and provide a useful model for in-depth study of development,the mechanism of evolution of oral verrucous carcinoma(ovc).Materials and Methods:Balb/c nude mice were used in this study. The sample of ovc was implanted into right anterior back and right posterior back of the mice with the methods of tissue transplantation and cell suspension,to establish model of ovc in nude mice (model1).The sample of postoperative recurrent oscc with metastatic cervical squamous cell carcinoma(scc)was implanted into the mice with the methods of tissue transplantation,to establish model of postoperative recurrent oscc with metastatic cervical squamous cell carcinoma(scc) models(model 2).To observative tumor growth characteristions of xenotransplanted tumor,success rate of xenotransplanted tumor,rendered growth curve and have a pathological examination of xenotransplanted tumor.Results:Model 1 has not obtained succeed,the success rate of model 2 is 87.5%(7/8),the tumor growth curve is to accord with the general law of the Gompertz function.There was necrosis in all of the tumors finally,and some had regressed.The xenotransplanted tumor was round or ellipsoidal shape with intact membrane and smoot surface.Occasionally,the surface of tumor was nodular.Their activitie were good,some were concentric necrosis,and some capillaries could be seen clearly on the surface of the tumor,a fish-shaped profile,with some kind of white-bean dregs.The cross section of tumor was solid and gray-white color,liked as fish.Light microscope(LM):The result revealed that the transplanted tumor was arranged in mass.The tumor cells were round or multisided.The cytoplasm of cell was stained red.The nuclei were round and stayed in centre.The proportion of the nuclei and cytoplasm was imbalance.Nuclear division could be easily observed.There are no transfer in Liver,lung and forelimb armpits and groin lymph nodes werereactive hyperpla.There was no difference of the expression of survivin and Bcl-2(B-cell lymphoma/leuke-mia-2) between wd-OSCC and xenograft tumor(P＞0.05).Conclusion:1 The nude mice Primary xenografts model of postoperative recurrent oscc with metastatic cervical squamous cell carcinoma(scc) model has been established.It laid a solid foundation for the establishment of a stable model of the transplant.2.The transplanted tumor maintained the properties of human oral squamous cell carcinoma. 3.There was no difference of the expression of survivin and Bcl-2 between wd-OSCC and xenograft tumor.
