Objective:In the experiment, human cervical carcinoma Hela229 cells were inoculated subcutaneously into nude mouse model to establish the xenografted tumor nude mice model about cervical cancer.Made Rhizoma Zedoariae affect the xenografted tumor and observed the effect of Rhizoma Zedoariae on growth of xenografted tumor model, to provide the theoretical basis for further research and clinical treatment of cervical cancer.Methods:Human cervical cancer Hela229 cells cultured in vitro, establish the model of Balb/c-nu transplantation tumor in nude mice cervical cancer 50, divided into 5 groups, 10 rats in each group. Respectively, the negative control group: normal saline(NS, 10ml/kg), the next day by gavage for 1 times; the positive control group: cisplatin(DDP, 25mg/kg), after intraperitoneal injection, every other day for 1 times; low dose, middle dose of Rhizoma Zedoariae, high dosage treatment group: the doses were 30, 60, 120mg/kg, the alternate day intragastric administration of 1 times, 10 times. General observation of each group of nude mice, body weight and tumor volume every 5 day measurement in nude mice. After the end of the experiment were sacrificed, remove the tumor, called tumor body weight, calculate the inhibition rate of tumor growth, HE staining to observe the pathological changes of each xenograft in nude mice.Results:1.Rhizoma Zedoariae turmeric in all treatment groups in nude mice had good mental state, diet, activity and reaction had no obvious change, weight does not appear significantly reduced, there is no significant difference between the weight of nude mice and saline group(P>0.05); cisplatin in mice of the control group gradually appear listlessness, activity weakened, slow response, poor appetite, weight loss, there is obvious the difference between weight changes and physiological saline control group(P<0.001).2.Different doses of Rhizoma Zedoariae turmeric group tumor volume with saline group with a significant difference(P<0.001); Compare the effect of transplantation tumor volume Curcuma between groups, the difference was statistically significant(P<0.05); The dose of Curcuma turmeric group and cisplatin in comparison to the control group, the difference was statistically significant(P<0.05).3.Low dose, middle dose and high dose group and the control group of Rhizoma Zedoariae turmeric cisplatin tumor growth inhibition rates were 41.39%, 61.92%, 76.28% and 63.61%, compared with the saline control group, there was significant difference(P<0.05); effect of Rhizoma Zedoariae on transplanted tumor weight between groups were compared, the difference was statistically significant(P<0.05); And the middle dose of Rhizoma Zedoariae turmeric group and cisplatin in comparison to the control group, no statistical difference(P>0.05).4.In the pathological sections visible Rhizoma Zedoariae treatment group tumor tissue shows patchy, punctate necrosis, structure of necrotic cells with hyperchromatic nuclei is not clear, visible at the same time, smaller cell apoptosis, nuclear pyknosis, irregular type.The saline group was seen under the microscope showed typical nuclear transplantation tumor of large deep staining, the cells are large, arranged in disorder, non polarity, complete capsule, pellets, plate-like, a small amount of lymphocytes and neutrophils infiltration.Conclusion:Rhizoma Zedoariae may have effects on inhibiting the proliferation of cervical cancer cells and promoting cell apoptosis, with a dose-dependent manner that as the dose increased, the antitumor effect of strengthened gradually. Its injury to the body and the side effect is not obvious, it has more safety and certain guiding significance to the clinical guide for the treatment of cervical cancer.
