Inhibitory effect of IL-24combined with DDP on Cervical Cancer in nude mice and its relationship with MTA-1expressionPurposeTo investigate the inhibitory effect of gene recombinant plasmid pDC316-hIL-24used alone or in combination with DDP on human cervical cancer nude mice model tumor growth as well as lymphatic metastasis, and its relationship with the expression of PDGF-B.MethodNude mice xenograft models were established with human cervical cancer Hela cells, and then animal models were randomly divided into six groups, each group had6mice:PBS (group A), empty plasmids(group B), recombinant plasmids pDC316-hIL-24(group C), full dose DDP(group D),recombinant plasmids pDC316-hIL-24+DDP(group E)as well as half dose DDP(group F).During the period of treatment, the tumor growth and the weight of nude mice models was monitored.At the end of the trial,the tumor was removed, the volume and weight of tumor was measured as well as found lymphatic metastasis, tumor growth inhibitory rate and metastasis rate was calculated.The expression of IL-24in transplanted tumor was identified by RT-PCR, observation of morphological changes of tumor tissues in each group by HE staining,PDGF-B was detected with Western blot and immunolhistochemistry technology.ResultpDC316-hIL-24expressed stably in Nude mice xenograft models, ts function of inhibiting tumor growth was significantly.The inhibitive rates of each groups were A:0B:4.3%、C:46.5%、D:52.1、E:42.3%、F:71.9%, respectively.The tumor inhibition rate of IL-24+DDP group higher than anther groups significantly(P<0.001).IL-24+DDP group has lower side effect of than DDP group, decline of weight was less,There was a significant difference in both groups(P<0.001).Pathological observation shows in plasmid IL-24group cervical cancer cells have focal necrosis and cells become large, and loose cytoplasm, nucleus deeply stained, pyknosis, nuclear dissolution, nuclear fragmentation.The result s of Western blot and immunolhistochemistry technology showed that the expressions of PDGF-B were significantly lower between IL-24group and IL-24+DDP group (P<0.001)。ConclusionThe combined intervention of IL-24and DDP on human cervical neoplasm nude mice model has significant inhibiting growth function, less side effect compared with other groups,and its can reduce lymphatic metastasis rate. The effect is likely associated with the specific down-regulated expressions of PDGF-B.
