Objective:To observe the effects of BTY on interleuki-6(IL-6)in serum in experimental osteoarthritis of rabbit,and investigate the mechanism of BTY on prevention and treatment of OA.Method:The healthy rabbits of new Zealand aged of 4-6 six months,half male and half female,were randomly collected 6 as the normal control group (Group A),Other 36 rabbits were construct the osteoarthritis model of rabbit with Hulth technique.After 8 weeks of operation,we confirm the building of the osteoarthritis model is successful by the alter of X-ray and pathology.Then they were randomly divided into five groups:the model control group(B group),the positive control gruop(C group)(glucosamine sulphate 35mg·(kg·d)-1),BTY high-dose group(Dh group)(120 mg·(kg·d)-1),midst-dose group(Dm group)(60 mg·(kg·d)-1)and low-dose group(Dl group) (30 mg·(kg·d)-1).There are 6 rabbits in each groups,And giving drugs by pouring stomach in enactment doseage.45 days later,we collect the blood of everyone rabbits to detect the level of IL-6 in serum,and then kill the rabbits and cutting the specimen of thighbone cartilage.Taking the specimen dehydration,embedding the specimen in paraffin and slice up.To dry with H.E. We grade the tectology with the change of articluar cartilage by the methods of Wakitani under the microscope on the way of double blind.And we make the correlation analysis with the Wakitani method learn the grade point index number and the level of IL-6 in serum.Result:(1)build the joint pathology of the modal animal to change to match the pathologic changes of the OA,and the appearance of its Wakitani method learn the grade point index number to 11.67±0.52,but normal animal was 0,and its difference have the high degree obviously,P<0.01;(2)The appearance of Wakitani method learn the grade point index number in group C,Dh,Dm,Dl were significantly lower than that of group B (6.68±0.74,3.29±0.49,4.50±0.55,7.83±0.98,11.67±0.52,P<0.01), showing a dose-dependent inhibiting effect.among them,the effect strength in group Dh and Dm is superior to that of group C.(3)Great significant contrast(P<0.01)were found between the level of IL-6 in serum in model group(173.1±72 pg/ml)and normal group(42.3±17.2 pg/ml);the level of IL-6 in serum in group Dh,Dm and C were significantly lower than that of group B(P<0.05),showing a dose-dependent inhibiting effect.among them,the effect strength in group Dh is superior to that of group C.(4)Pearson's correlation analysis showed that the level of Il-6 in serum was positively correlated with the appearance of its Wakitani method learn the grade point index number and the correlation coefficient was 0.98.Couelusion:(1)BTY can decrease the level of IL-6 in serum of experimental rabbits OA,its strength showing a dose-dependent inhibiting effect,and among them,the effect strength of High-dose BTY is superior to glucosamine sulphate in decreasing the level of IL-6 in serum in experimental rabbits OA.(2)BTY to inhibite the level of IL-6 in serum with appearance of Wakitani method learn the grade point index number was examined by Pearson's correlation analysis and was positively related.
