Objective:To study the combined effects of glucosamine hydrochloride Ibuprofen different stages of osteoarthritis in rabbits, and to explore the mechanism of combination therapy, and provide data for clinical research.Methods:84rabbits were randomly divided into seven groups (n=84):sham operation group (A), model group (group B), and right in the ibuprofen group (Group C), glucosamine hydrochloride group (D group), compound high group (E), compound of group (F), compound low group (G group), B, C, D, E, F, G animals were carried out on both sides of the knee joint modeling. Anesthesia is the ear vein injection of20%urethane (5ml/kg), under sterile conditions in the side of the patella knee beside the incision, isolated medial collateral ligament, the medial collateral ligament and then cut off about0.5cm length; total removal of the media! meniscus, wound closure. A group of modeling the same conditions as the other group, after just cut the joint cavity, without any treatment, suture joint cavity and skin. After modeling, each animal intramuscularly400,000units of penicillin to prevent infections, penicillin injections three days in a row. Group C orally administered25mg/kg dexibuprofen solution, D group60mg/kg orally administered glucosamine hydrochloride, E group gavage25+60mg/kg of compound solution, F group gavage12.5+30mg/kg Compound solution, G group gavage6.25+15mg/kg of compound solution A, group B ig0.5%CMC solution. Continuous administration of120days, the animals were sacrificed at30,120days (30days in each group were sacrificed4,120in each group were sacrificed on day8), carried rabbit knee Gotdbrg VM score; detection of serum IL-1(3, TNF-a, ALP level content; synovial fluid chondroitin sulfate, measuring phospholipase A2activity; articular cartilage reasonable inspection.Results:Rabbit knee Gotdbrg VM score showed that administration of30days and120days, A, group B compared to the indicators showed significant differences (p<0.05), the treatment group compared with the group B of each index, there was a significant the difference (p<0.05).30-day dosing, serum IL-1β, TNF-a levels in group B was significantly higher in group A, ALP levels in group B was significantly lower in group A, there was a significant difference (p<0.05), treatment group after administration serum IL-1β, TNF-a levels were significantly lower than in group B, ALP levels were significantly higher in group B, there was a significant difference (p<0.05); chondroitin sulfate levels in synovial fluid were significantly lower than in group B A group of phospholipase A2group B was significantly higher than that in group A, there was a significant difference (p<0.05), treatment group after administration of chondroitin sulfate in synovial fluid were significantly higher than group B, phospholipase A2levels were significantly lower than group B, there was a significant difference (p<0.05).120days articular cartilage HE staining showed that compared with the model group, C, D, G group articular cartilage injury score was no significant difference, E, F group with a significant difference (p<0.05).Conclusion:Ibuprofen joint OA glucosamine hydrochloride inhibits the IL-1β, TNF-a levels, reduce the expression of phospholipase A2activity increased in OA disease ALP, chondroitin sulfate levels, thus improving symptoms of OA, promote cartilage cell regeneration, reducing the destruction of cartilage, and thus delay the development of osteoarthritis.
