Objective:To investigate the damaging effect and mechanism of eating high fat/high sucrose on degeneratice osteoarthritis in rabbit.Methods:Male New Zealand white rabbits were divided into four groups:normal fodder group(NG,n=10)and high-sugar and high-fat fodder group(SFG,n=10,with 10%lard,37%white cane sugar and 53% normal fodder),high-sugar fodder group(SG,n=10,with 37%white cane sugarand 63%),high-fat fodder group(FG,n=10,with 20%lard,80% normal fodder),and observed for 28 weeks.Once a month after a overnight fasting the blood samples of the rab biIs were tested for the levels of blood glucose,insulin,and trigly- ceride.At the end of 28 weeks all the rabbits were killed to get the knee articular cartilage,which were stained with H.E,and then observed under microscope.A histologic evaluation was done according to the modified Mankin grading scale and thickness of cartilage and subchondral bone respectively.Another part of pancreas was fixed with 2.5%glutarald- ehyde,sampled by means of normal electron microscopy,and observed and photographed under transmit electron microscope.Results:(1)Compare with four groups,the level of blood insulin in FG(1.79±0.89)and SFG(1.97±0.90)was significantly higher than NG(0.57±0.16)and SF(0.60±0.19)after one month(P<0.05),and then, decreased gragually.Finally,it sustained in a low-grade level for at least three months,the level of blood insulin in SG increased more slowly than that in FG and SFG.It achieved to a peak after three month,then decreased gragually.And the level of weight in SG was obviously higher than that in NG(P<0.05).FBG,TC,TG in experiment groups were obviously higher than that in NG,espcially in that of SFG.(2)By macroscopic observation and microscopic observation,artilage degeneration in the FG and SFG was significantly cataplasis than that in the other groups. (3)Mank- in scors and the thickness of cartilage in FG and SFG were significiantly higher than that in the other two groups(P<0.05).The thickness of subchondral bone in FG and SFG were higher than that in NG(P<0.05).Conclusion:The results suggest that eating adipose food and high-sugar and high-fat fodder diet maybe induce or aggravate the disorder of articular cartilage.
