Objective:To study the effect of maternal high-sucrose diet on the glucose metabolism of offspring rats,and its programming effect.Methods:The experiment divided the Sprague-Dawley offspr:ing rats into maternal high glucose drinking water and normal drinking water group,on this basis,the group of four months of male offspring were divided into two groups,one group was the offspring of normal drinking water,the other group was high-sucrose drinking water group until 6 months old.To this end,there are 4 groups in this experiment:maternal and offspring normal drinking group(CC),maternal normal drinking and offspring high-sucrose drinking group(CH),maternal high-sucrose drinking and offspring normal drinking group(HC),maternal and offspring high-sucrose drinking group(HH).When the offspring are 6 months,detections were followed aspects:1)Growth curve;2)Daily intake of diets,drinking water and total calorie;3)Fasting plasma glucose,fasting insulin and glucose tolerance;4)Observation of islet structure changes under light microscope;5)The expression of IR,IRS-1 and Akt was detected by qPCR;6)Western blot was used to detect the phosphorylation level of the insulin receptor substrate-I serine residue(p-IRS-1 S612)and Akt phosphorylation activation(p-Akt)levels.Results:The birth weight of the offspring exposed to high-sucrose during pregnancy increased significantly,this difference disappeared after 3 weeks of birth.,After adult offspring to give high-sucrose drinking water(CH and HH groups)its weight gain,fasting plasma glucose increased,glucose tolerance reduced,insulin sensitive decline and triglyceride levels increased,compared with adults who maintain normal drinking water after adulthood.But there was no significant difference between the two groups.At the mRNA level,the expression of IR,IRS-1 and Akt were decreased in HC,CH and HH groups.The expression of p-IRS-1 S612 was significantly different in CH group and HH group and p-Akt had no significant difference between the four groups.Conclusion:Maternal high-sucrose drinking can affect the glucose metabolism of adult offspring rats,regardless of adult exposure to high sugar environment.The adverse effects of pregnancy and adult offspring exposed to the same high-sucrose drinking were not significantly expanded,compared with the offspring of normal water during pregnancy.
