ObjectiveTo evaluate the expression of HIF-1αand PTEN in epithelial ovarian cancer,and to explore the relationship between their expression and clinicopathological characteristics.Then to analysis the relationship between HIF-1αand PTEN in the development of epithelial ovarian cancer in order to take a new way on guiding clinical diagnosis and treatment on early stage ovary cancer and recurrence ovary cancer.MethodsThe expressions of HIF-1αand PTEN in 10 of normal ovarian tissue,15 of benign ovarian tumors,12 of borderline epithelial ovarian tumors and 32 of epithelial ovarian carcinoma were examined by S-P immunohistochemistry.All data were analyzed using statistic software SPSS 13.0.The correlations of HIF-1αand PTEN with clinicopathological factors were analyzed by x~2 test or Fisher's exact test;their relationship by Spearman;The criterion of test was a=0.05.Results1.There was no expression of HIF-1αin normal ovarian tissue.The overexpression of HIF-1αwas higher in epithelial ovarian carcinoma than in borderline epithelial ovarian tumors as well as higher in borderline epithelial ovarian tumors than in benign ovarian tumors,the difference was significant(P<0.05).The overexpression of HIF-1αin orderline epithelial ovarian tumors and epithelial ovarian carcinoma were significantly higher than that in normal ovarian tissue and benign ovarian tumors(P<0.05);but the expression of HIF-1αfrom orderline epithelial ovarian tumors to epithelial ovarian carcinoma was not significant difference(P>0.05).2.In ovarian carcinoma group,no correlation was found between the expression of HIF-1αand age,histological subtype,FIGO stage,lymph node metastasis(P>0.05);but The overexpression of HIF-1αin poorly differentiated group was significantly higher than in well differentiated group,the difference was significant(P<0.05). 3.The expression of PTEN in normal ovarian tissue and benign ovarian tumors was significantly higher than that in epithelial ovarian carcinoma(P<0.05); the difference of PTEN expression between in pithelial ovarian carcinoma and in borderline epithelial ovarian tumorswas was significant(P<0.05).The expression of PTEN was higher in normal ovarian tissue than in benign ovarian tumors as well as higher than in borderline epithelial ovarian tumors,but the difference was not significant(P>0.05).4.In ovarian carcinoma group,no correlation was found between the expression of PTEN and age,histological subtype(P>0.05).The expression of PTEN in well differentiated group was significantly higher than in poorly differentiated group(P<0.05).The expression of PTEN in FIGOⅠ,Ⅱstage or no lymph node metastasis group was significantly higher than in FIGOⅢ,Ⅳstage or lymph node metastasis group(P<0.05).5.In ovarian carcinoma group,there was negative correlation between the expression of HIF-1αand expression of PTEN(r_s=-0.629,P=0.000).Conclusions1.With the development of epithelial ovaian tumors from benign to malignant, HIF-1αexpression increases,which indicates overexpression of HIF-1αappears in the early stage of ovarian epithelial cancer,and may play an important role in the occurence of ovarian cancer.2.In epithelial ovarian carcinomas,the expression of HIF-1αhas positive correlation with histological grades.It could indicate that HIF-1αplay a key role in initiation and progression of epithelial ovarian carcinomas.3.Elevated expression of PTEN was observed in normal ovarian tissue and in benign ovarian tumors,but there was no expression of PTEN in epithelial ovarian carcinoma.It suggested that expression of PTEN may play a role in the carcinogenesis of ovarian carcinoma.4.In epithelial ovarian carcinomas,the expression of PTEN was correlated with FIGO stage,lymph node metastasisand grade.It suggested that of negative expression of PTEN may make it easier for epithelial ovarian carcinoma to metastases,which cause poor prognosis. 5.In epithelial ovarian carcinomas,there was negative correlation between the expression of HIF-1αand expression of PTEN,which suggested that expression of HIF-1αmay be in coordinated with negative expression of PTEN in carcinogenesis and progression of epithelial ovarian carcinoma.
