Aromatase (also called aromatase cytochrome P450, P-450arom) is the important enzyme in the production of estrogen. Estrogen combined with estrogen receptor (ER) can produce effect. Progestogen receptor (PR) must be produced by effect of estrogen. Therefore aromatase ER and PR all have close relation with estrogen. This experiment investigate distributions and expressions of aromatase ER and PR in normal ovarian tissues ovarian serous papillary cysta-denoma and ovarian serous papillary cystadenocarcinoma to study the relationship between these factors and the pathogenesis clinic stages and histology types of ovarian serous papillary cystadenocarcinoma, to study whether aromatase have the correlation with ER and PR and to investigate their clinic significances.Materials and Methods1. Materials: Paraffin - embedded tissues from 36 patients with ovarian serous papillary cystadenocarcinoma by way of experiment group obtained from patients who received surgery at the affiliated hospital of China Medical University between 1995 -2002 ,they had full data of pathology, had not received either chemotherapy or radiation therapy before or after surgery, patients with a mean age of 47. 75(rangel7 -67). 10,13 and 13 of the cases were classified as to histological differentiation well - differentiated moderately differentiated and poorly differentiated. 3, 8, 20 and 5of the cases were respectively classified as to FIGO stage I , II , III, IV. At one time paraffin - embedded tissues from 10 patients with normal ovarian tissues and 20 patients with ovarian serous papillary cystadenoma by way of comparison group, patients with a mean age of 26. 4(range17 -42) and 47. 2(range40 -53). Possession pathology substantiated by the pathologic expert , clinic by stages according as ovarian malignancy standard by FIGO 1986.2. Methods: Immunohistochemistry SABC method was used to investigate the distribution of aromatase . All specimens were fixed in 4% buffered formaldehyle and prossed into routinely paraffin - embedded blocks. For each case, five - micrometer sections from these blocks were mounted on silane - coated slides, air-dried. Sections were dewatered and hydrated, and then treated in 3%hydrogen peroxide for 10 min to quench endoengous peroxidase. Prior to staining with the anti - P450arom antibodies (1:300 ) , heat antigen - rehabilitate technology was used, and then non - immuno animal serum was droped to seal. All procedures were performed at room temperature and in a moist chamker, with a PBS wash after each step . P450arom antibody were droped on the slides. After over incubation at 4C , the sections were incubated with then counter - stained in Mayer' s heamatoxylin and mounted with Crystal mont . Immunohistochemistry two steps method to investigate the distribution condition of ER PR . namely to heat antigen - rehabilitate technology was used, After over incubation at 4C , wash with PBS , apply one drop of reagent B , incubate at room temperature for 30 minutes , wash with PBS, incubate with DAB , counterstain with hematoxy-lin and mount with coverslip . Control sections with negative expression were incubated with PBS, we used known cancer sections as control group with positive expression.3. Statistical methods; Statistical analyses of protein expression was carried out the Chi - square method by SSPS 10.0. Differences were considered significant when P values were less than 0.05.Result1. The expression of aromatase in the three kind of tissue was mostly located at cell plasma, The expression of ER and PR in the three kind of tissue was mostly located at cell nuclear.2. The positive expression rates of aromatase were 30% 40% 69% in nor-mal ovarian tissues and in ovarian serous papillary cystadenoma and in ovarian serous papillary cystadenocarcinoma. The positive expression rates of aromatase in ovarian serous papillary cystadenocarcinoma were higher than those in normal and ovarian serous papillary cystadenoma ( P <0. 05).
